目的建立高效液相色谱法测定蛋黄油中胆固醇的含量。方法样品无须皂化处理,直接用二氯甲烷-甲醇(1∶2)溶解,经反相C18色谱柱(150 mm×3.9 mm,5μm)分离后测定,以甲醇为流动相A,以甲醇-异丙醇-正已烷(10∶70∶20)为流动相B;梯度洗脱;检测波长为205 nm;流速为1.0 m L/min;柱温为35℃。结果本方法测定胆固醇线性范围为0.097 23~1.94463 mg/m L,线性方程y=3 869.446x+15.519,相关系数r=0.999 9,线性关系良好;加样回收率为96.98%~100.4%。结论该方法灵敏、快速、准确,适用于测定蛋黄油中胆固醇的含量。
OBJECTIVE To establish a method for determination of Cholesterol in egg yolk oil by HPLC. METHODS The sample was dissolved in Dichloromethane-Methanol( 1∶ 2) without saponification. The chromatographic separation was carried out on a C18( Waters C18: 150 mm × 3. 9 mm,5 μm) column using UV detector at 205 nm. Methanol and Methanol-Isopropyl alcohol-n-hexane( 10∶ 70 ∶ 20) were used as mobile phase by gradient elution. The flow rate was 1. 0 m L/min,column temperature was 35 ℃.RESULTS The linear range of Cholesterol was 0. 097 23- 1. 944 63 mg / m L,Linear equation was y = 3 869. 446 x + 15. 519( r = 0.999 9). The average recovery range was 96. 98%- 100. 4%. CONCLUSION The method is simple,quick and accurate. It can be used for determination of Cholesterol in egg yolk oil.
