目的探讨丹参酮ⅡA对人肝癌细胞系SMMC-7721生长活性及对奥沙利铂敏感性的影响和机制。方法不同浓度的丹参酮ⅡA和奥沙利铂单独或联合作用人肝癌细胞系SMMC-7721 72 h后,采用噻唑蓝(MTT)法检测该细胞系的生长活性,分析丹参酮ⅡA与奥沙利铂在抑制肝癌细胞增殖中的相互作用。流式细胞术分析丹参酮ⅡA单独及联合应用奥沙利铂对肝癌细胞凋亡的影响。结果实验所选各种浓度丹参酮ⅡA对SMMC-7721均表现出一定程度的生长抑制作用,并呈剂量依赖性;当丹参酮ⅡA质量浓度为1.0,2.0,5.0,10.0 mg·L^-1时,合用质量浓度为1.0,2.0,5.0 mg·L^-1的奥沙利铂能显著抑制SMMC-7721细胞的增殖,并显示出两种药物的协同作用(q〉1.15)。流式细胞术分析发现两种药物均可有效诱导SMMC-7721细胞的凋亡,且在上述浓度联合应用时具协同效应。结论丹参酮ⅡA可以显著抑制肝癌细胞系SMMC7721细胞增殖,并能增强该细胞系对奥沙利铂的敏感性,其作用机制可能与丹参酮ⅡA和奥沙利铂协同诱导细胞凋亡有关。
OBJECTIVE To investigate the effects of tanshinone Ⅱ A on the growth activities and oxaliplatin sensitivity in human hepatocellular carcinoma cell line SMMC-7721 and its mechanism. METHODS After treating SMMC-7721 cells with different concentrations of tanshinone Ⅱ A with or without oxaliplatin for 72 h, MTT assay was used to examine the growth activities on these cells. The cooperative effects of tanshinone Ⅱ A and Oxaliplatin on inhibiting the growth of SMMC-7721 cells were analyzed. Flow cytometry (FCM) was used to examine the effects of Tanshinone Ⅱ A alone and Tanshione combinated with oxaliplatin on the apoptosis of SMMC- 7721 cells. RESULTS Tanshinone Ⅱ A produced concentration-dependented antiproliferative effect on SMMC-7721 cells at all experimental concentrations. Enhanced antiproliferative effects were obtained when SMMC-7721 cells were cultured with tanshinone ⅡA at 1.0,2.0,5.0,10.0 mg · L^-1 combined with 1.0,2.0,5.0 mg· L^-1 oxaliplatin (q 〉 1.15). FCM results indicated that the apoptosis was induced effectively by both tanshinone Ⅱ A and oxaliplatin in SMMC-7721 cell, and synergic effect at above-mentioned concentrations was found in inducing SMMC-7721 cells apoptosis. CONCLUSION Tanshinone Ⅱ A can significantly inhibit the growth of SMMC-7721 cells, and enhance its oxaliplatin sensitivity, the mechanism may be associated with the synergic effect of tanshinone Ⅱ A and oxaliplatin in inducing apoptosis.
