目的:观察受体酪氨酸激酶c-kit及其配体干细胞因子(stem cell factor,SCF)在人正常肝、肝硬化、癌旁肝及肝癌组织中的共表达。方法:用免疫组化/免疫荧光染色方法和组织芯片检测受体酪氨酸激酶c-kit及其配体在166例肝癌和配对癌旁肝、16例肝硬化及10例正常肝组织中的表达差异及其共表达状况。结果:免疫组化检测c-kit在肝癌、癌旁肝、硬化肝和正常肝组织中的阳性率分别为47.59%,95.78%,93.75%和100%,其中强阳性(≥++)率分别为27.71%,77.71%,87.53%和100%,c-kit蛋白在肝癌组织中明显低表达;而SCF在肝癌、癌旁肝、硬化肝和正常肝组织中的阳性率分别为68.07%,100%,100%和100%,其中强阳性(≥++)率分别为35.54%,94.58%,93.75%和100%,SCF在肝癌组织中也呈现低表达;但SCF在各种组织中的表达均高于c-kit。免疫荧光检测受体酪氨酸激酶c-kit及其配体SCF在各种组织中均有不同程度的共表达,且均定位于肝癌细胞和肝细胞的细胞质中,其中在肝癌、癌旁肝、硬化肝和正常肝组织中的共表达率分别为51.20%,100%,100%和100%,而肝癌组织的强阳性共表达(≥++)率仅为25.30%,其他组织均高于95%。结论:与正常肝、硬化肝、癌旁肝组织相比,肝癌组织中c-kit和SCF均呈低表达,但SCF在相应组织中的表达稍高于c-kit;正常肝、硬化肝、癌旁肝组织中c-kit/SCF共表达远高于肝癌组织。
Objectives: To evaluate the co-expression of the receptor tyrosine kinase c-kit and its ligand stem cell factor (SCF) in tissues from human normal liver, cirrhotic liver, noncancerous liver, and hepatocellular carcinoma (HCC) . Methods: The differential expression patterns and co-expression of c-kit and SCF were examined by immunochemical and immunofluorescent method on a tissue microarray, including samples of 10 human normal livers, 16 cirrhotic livers, 166 HCC, and matched adjacent noncancerous liver tissues. Results: Immunochemical analysis showed that positive rates of c-kit staining were 47. 59%, 95. 78%,93. 75%, and 100% in hepatocellular carcinoma, adjacent noncancerous liver, cirrhotic liver, and all of human normal liver tissues, respectively. The corresponding strong positive(≥++) ratios were 27.7%, 77.7%, 87.5%, and 100.0% ,respectively. The positive rates of SCF were 68.1%, 100%, 100 %, and 100% in HCC, the adjacent noncancerous liver, cirrhotic liver, and human normal liver tissues, respectively. The corresponding strong positive(≥ + +) ratios were 35.54 %, 94.58 %, 93.75 %, and 100%, respectively. The expression of SCF was lower in HCC. But the expression of SCF was higher than c-kit in all tissues. Immunofluorescent analysis showed co-expression of c-kit and SCF in various tissues at different degrees. They were localized in the cytoplasm of HCC and normal liver. The co expression rates were 51.20% ,100% ,100%, and 100% in HCC, the adjacent noncancerous liver, cirrhotic liver,and human normal livor tissues, respectively. The strong positive(≥+ +) ratio of co expression was only 25.30 % in HCC and more than 95 % in other three tissues. Conclusion: The expressions of c-kit and its ligand SCF are lower in HCC compared with normal liver, cirrhotic livers, and adjacent noncancerous liver tissues.
