目的建立一种简易、高效的小鼠2/3肝切除方法。方法取3月龄健康昆明小鼠,进行肝顺次结扎切除手术,观察术后小鼠生存和肝组织再生状况。结果通过顺次结扎切除左小叶和中央小叶,可在15min内完成小鼠2/3肝脏切除手术,术后成活率为90%,术后42h时可见肝组织再生,术后7d肝脏可恢复75%以上的肝组织原质量。结论通过分叶顺次肝切除术,可准确量化肝脏切除的程度,简便易行、成功率高,为肝再生的机理研究提供了理想的动物模型。
Objective To explore an effective and convenient method for 2/3 partial hepatectomy in mice.Methods3-month old Kunming mice were anesthetized by 2.5% 2,2,2-Tribromoethanol anesthetic(0.1 mL/10 g intraperitoneal injection).To remove 2/3 liver,a larger abdominal incision(about 3 cm) was made and the right as well as the central hepatic lobes were resected separately.After surgery,the liver regeneration status and the mice survival conditions were investigated.Results The morphology and ratio of the hepatic lobes were constant.By resecting the right and the central hepatic lobes in succession,we successfully did the 2/3 partial hepatectomy in mice.The surgery could be finished within 15 minutes and the postoperative survival rate was 90%.The remained 1/3 liver could recover to 75% original liver weight during 7 days liver regeneration and almost 100% original liver weight after 15 days.The active proliferation status of hepatocytes was observed in the remained 1/3 hepatic lobes by HE histopathological assay.Conclusion We successfully established a 2/3 partial hepatectomy method in mice.This method could control the liver resection accurately,conveniently and with high success rate.Our experiments offered an ideal animal model for the liver regeneration study.
