目的探讨β-七叶皂甙钠对脑外伤小鼠脑皮质及海马AQP9水通道蛋白9(AQP9)表达的影响。方法昆明小鼠120只,随机分为假手术组(40只)、脑外伤组(40只)和β-七叶皂甙钠组(40只)。采用Feeney自由落体法制作脑外伤模型,采用干湿重法测定脑含水量,免疫组化和Western blot方法检测各组小鼠大脑皮质及海马水通道蛋白9的表达。结果相比于对照组,脑外伤后3h,小鼠脑含水量明显上升,6h继续升高,24h达峰值,3d时开始下降;相比于脑外伤组,相同时间点的β-七叶皂甙钠组小鼠含水量则明显降低(P〈0.05)。免疫组化和Western blot结果显示,相比于对照组,脑外伤后3h,大脑皮质及海马AQP9的含量明显增高,6h后略有降低,24h后再次明显增高,并于3d时达峰值;相比于脑外伤组,相同时间点的β-七叶皂甙钠组小鼠大脑皮质及海马AQP9的含量则明显降低(P〈0.05)。结论β-七叶皂甙钠能够通过降低AQP9蛋白表达参与脑外伤水肿的调节。
Objective To study the effect of β-sodium aescinate on the expression of aquaporin-9 in the cortex and hippocampus of mice after traumatic brain injury. Methods 120 Kunming mice were randomly divided into the sham group(40), traumatic brain injury group(40) andβ-sodium aescinate group(40). The traumatic brain injury model was established according to Feeney's method.Dry-wet weight method were used to measure the extent of brain edema.The expression of aquaporin 9 in the cortex and hippocampus of mice was detected by immunohistochemistry and Western blot methods. Results Compared with the sham group, the brain water content was increased significantly at 3h, continues to rise at 6h, reached peak at 24 h, began to drop on 3d in the cortex and hippocampus of mice(P〈0.05),but was decreased significantly in celecoxib group than in traumatic brain injury group(P〈0.05) at the same time point. Compared with the sham group, the expression level of AQP9 protein was increased significantly at 3h, slightly decreased at 6h, significantly increased at 24 h again, reached the peak at 3d(P〈0.05), however, was decreased significantly in celecoxib group than in traumatic brain injury group(P〈0.05) at the same time point. Conclusion The reducing edema of traumatic brain injury byβ-sodium aescinate might be related to the downregulation of AQP9 expression.
