目的探讨单唾液酸神经节苷脂(GM1)对脑缺血再灌注大鼠海马ARMS与CrkL表达的影响。方法96只SD大鼠随机分为:假手术组(Sham,32只)、神经节苷脂治疗组(GM1,32只)、缺血再灌注组(I/R,32只)。GM1治疗组和I/R组依据缺血后再灌注的不同时间点再细分为6 h、12h、24 h、3 d 4个组。应用免疫组织化学方法和Western blot方法检测各组大鼠海马ARMS与CrkL蛋白的表达。结果免疫组织化学方法及Western blot检测发现,假手术组大鼠海马有微量ARMS与CrkL表达,而经过脑缺血再灌注处理后,ARMS与CrkL表达水平在6 h已显著升高,在24h时达到峰值最高,3d时表达较假手术组仍显著升高;与同一时间点的缺血再灌注组大鼠比较,GM1治疗组的ARMS与CrkL蛋白阳性表达亦显著升高(P〈0.05)。结论GM1对脑缺血再灌注损伤的神经保护作用可能与上调ARMS与CrkL蛋白的表达相关。
Objective To study the of monosialoganglioside on the expression of ankyrin-rich membrane spanning protein(ARMS) and CrkL in hippoeampus of cerebral isehemia-repeffusiou rat. Methods 96 SD rats were randomly divided into: the sham group, ischemia-reperfusion group(I/R), monosialoganglioside group(GM1) at different time points (6h, 12h, 24h, 3d) on average. The expressions of ARMS--~CrkL in hippoeampus were examined by immunochemistry and Western blot methods. Results Western blot and immunochemistry showed that the positive expressions of ARMS and CrkL in hippoeampus were a little in sham group, but increased significantly at 6h after cerebral ischemia-reperfus(P〈0.05), reached the peak at 24h, and still higher significantly than in sham group at 3d. The positive expression levels of ARMS and CrkL in hippocampus were increased significantly in GM1 group than in I/R group at the same time points(P〈0.05). Conclusion The protective effect of monosialoganglioside the cerebral ischemia-reperfusion injury might be related to the up-regulation of ARMS and CrkL expressions.
