目的探讨重组人促红细胞生成素(rHuEPO)对脑缺血再灌注小鼠海马肿瘤坏死因子α(TNF-α)表达的影响。方法将96只昆明小鼠随机分为:假手术(sham)组、rHuEPO治疗组和缺血再灌注(I/R)组,每组32只。每组依据缺血后再灌注不同时间点再分为6 h、24 h、3 d及7 d 4个小组。应用免疫组化和Western blot检测各组小鼠海马TNF-α蛋白的表达,RT-PCR方法检测TNF-αmRNA表达的变化。结果免疫组化和Western blot检测结果发现,I/R组TNF-α蛋白表达量明显高于sham组和rHuEPO治疗组(P均〈0.05);RT-PCR结果发现,I/R组TNF-αmRNA表达量明显高于sham组和rHuEPO治疗组(P〈0.05)。结论rHuEPO可通过减少TNF-α的表达参与脑缺血再灌注损伤的神经保护作用。
Objective To study the expression of tumor necrosis factor-α(TNF-α) in cerebral ischemia-reperfusion mice after recombinant human erythropoietin(rHuEPO) was injected.Methods Ninety six Kunming mice were randomly divided into sham group,ischemia-reperfusion(I/R) group,rHuEPO group at different time points(6 h,24 h,3 d and 7 d) on average.The expression of TNF-α in hippocampus of all groups was examined by immunochemistry and Western blot methods.The expression of TNF-α mRNA was examined by RT-PCR method.Results The positive expression of TNF-α was increased significantly in I/R group than in sham group(P 〈 0.05),however the expression of TNF-α was lower in rHuEPO group than in I/R group(P 〈 0.05).The expression of TNF-α mRNA increased significantly in I/R group than in sham group(P 〈 0.05),however decreased in rHuEPO group.Conclusion rHuEPO has protective effect possibly through down-regulating the expression of TNF-α in cerebral ischemia-reperfusion injury.
