目的探讨T2DM患者血管内皮生长因子(VEGF)基因SNPrs3025039多态性。方法运用PCR-RFLP检测170例T2DM患者(T2DM组)和100名健康体检者(NC组)的VEGF基因SNPrs3025039多态性。结果 (1)两组的基因型和等位基因频率差异无统计学意义(P〉0.05),但糖尿病慢性肾脏疾病(CKD)组CT、TT基因型频率和T等位基因频率高于糖尿病非CKD(NCKD)组(P〈0.05);CC基因型和C等位基因频率低于NCKD组(P〈0.05);(2)T2DM组BMI、SBP、DBP、HbA1c、FPG、2hPG、TG水平高于NC组(P〈0.01),HDL-C水平较NC组降低(P〈0.01)。结合FPG、TG、TC、LDL-C及HDL-C进行多重Logistic回归分析显示,CT和TT基因型与CKD的发生呈正相关(OR=2.75,P〈0.05)。结论 T2DM患者VEGF基因SNPrs3025039多态性与正常人群比较差异无统计学意义,但CT、TT基因型与CKD的发病风险有关。
ObjeeUve To explore association of VEGF gene SNP rs3025039 polymorphism with development of T2DM. Methods VEGF gene polymorphism was detected in 170 T2DM patients and 100 controls by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results (1) Genotype and allele frequencies did not show significant differences between the controls and T2DM groups (P〉0. 05). Patients with chronic kidney disease in diabetes showed a higher frequency of the CT and TT genotype and T allele than those non-chronic kidney disease in diabetes; and a lower frequency of the CA3 and C allele than those non- nephropathy (P〈0. 05). (2)BMI, SBP, DBP, HbA1c, FPG, 2 hPG and TG in group of T2DM were significantly higher, and HDL-C was obviously lower than that of controls (P〈0. 01). The combination Of FPG, TG, TC, LDL-C and HDL-C for multiple logistic regression analysis showed that CT and TT genotype were positively correlated with the occurrence and development of chronic kidney disease in diabetes(OR=2. 75, P〈0. 05). Conclusion The difference of VEGF gene SNP rs3025039 polymorphism is not significant between the controls and T2DM patients, but CT and TT genotypes are positively correlated with the occurrence and development of T2DM.
