为测定及分析甘蔗在不同收获时间蔗糖含量的变化,采用水处理甘蔗样品,采取高效液相色谱法,经Waters Sugar Pak I柱(6.5mm×300mm,10.0μm),Waters2414示差折光检测器(RID)检测,流动相为高纯水,流速:1.0mL·min^-1,柱温:90℃,检测器温度:35℃.结果显示该法相对标准偏差(RSD)(n=6)〈2%,回收率为95%~105%,蔗糖质量浓度为10~50mg·mL^-1.且甘蔗在不同的收割时间蔗糖含量变化较明显,基本在14:00~15:00时达到最大值.实验表明HPLC法能够较快且较准确测出甘蔗中蔗糖的含量.通过比较可知,甘蔗蔗茎中蔗糖含量是随着光合作用的强弱而变化的.
In order to analysis and determination of sucrose in sugarcane reaping in different times. In the study, sampling water was used as solvent. HPLC is uesd, the chromatographic separation was achieved on an Waters Sugar Pak I column (6.5mm×300mm,10.0μm) , which was held at a constant temperature of 90 ℃ , RID temperature was set as 35 ℃, ultra-high purity water as moving phase, at a flow rate of 1.0 mL· min-1 The result shows average spike recoveries for 5 replicate determinations of three saccharides were found in a range of 95% to 105% , with a relative standard deviation (RSD) less than 2% , between 0. 89% and 0. 98%. And the concen- tration is between 10. 0 to 50. 0 mg · mL-1. Changes of sucrose in sugarcane reaping in different times are obviously, and generally reach their maximum at 14:00 to 15:00. The method of HPLC can quickly and accurately measures the content of sucrose in sugarcane. Through the comparison know sucrose content in sugarcane stalk is changes with the strength of the photosynthesis.
