目的 观察泰山照山白金丝桃苷(Hyperin)对人乳腺癌细胞株MCF-7细胞周期及凋亡影响及其机制.方法 体外培养人乳腺癌细胞MCF-7,加入不同金丝桃苷浓度处理,运用MTT法检测细胞生长、流式细胞术PI染色检测细胞周期,western blot检测细胞周期蛋白依赖性激酶2(cyclin-dependent kinase 2,CDK2)、细胞周期蛋白A (Cyclin A)及caspase3的表达.结果 与对照组相比,经金丝桃苷处理后细胞生长呈时间及浓度依赖性抑制;MCF-7细胞周期阻滞在G2/M期,该期细胞比例增多;CDK2、Cyclin A蛋白水平的表达与对照组相比明显下降而caspase3蛋白水平的表达明显升高.结论 通过细胞周期的G2/M期阻滞诱导细胞凋亡可能是金丝桃苷发挥抗肿瘤作用的可能机制之一.
Objective: To investigate the effect and mechanism of the Hyperin on cell cycle and apoptosis on human breast cancer cell lines MCF -7. Methods:MCF -7 ceils cultured in vitro, MTr assay was used to detect cell viability with various concentration of Hyperin, flow cytometry was used to detect cell cycle, western blot to test protein expression of cell cycle protein and apoptosis retelated protein. Results : Compared with vehicle treated, cell viability decreased in a dose and time dependent way treated with Hyperin, and cell cycle was blocked in G2/M phase and cell number in G2/M phase was increased and S phase was obviously decreased at the meantime, expression of cyclin - dependent kinase 2 ( CDK2), Cyc- lin A was decreased and caspase 3 were significantly increased compared to control group. Conclusion: By activating the cell cycle arrest related apoptosis pathway may be one of the mechanism of antitumor effect of Hyperin.
