目的研究莫诺苷对局灶性脑缺血再灌注大鼠纤维蛋白原(Fib)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)和凝血酶时间(TT)的影响。方法用线栓法制备大鼠局灶性脑缺血再灌注模型,造模后,分别给予莫诺苷低剂量(30mg/kg)、中剂量(90mg/kg)、高剂量(270mg/kg)连续灌胃7d,阿司匹林(ASA)为阳性对照药。使用试剂盒分别检测Fib、PT、APTT、TT指标的变化。结果与假手术组相比,模型组Fib含量显著增加(P〈0.001),PT、APTT、TT显著缩短(P〈0.001);与模型组相比,莫诺苷能明显降低Fib含量(P〈0.01),延长PT、APTT、TT(P〈0.05)。结论莫诺苷具有对抗局灶性脑缺血再灌注大鼠凝血功能的作用。
Objective To investigate the effects of morroniside on fibrinogen (Fib), prothrombin time (PT), activated partial thrombo- plastin time (APTT) and thrombin time (TT) in focal cerebral ischemia/reperfusion in rats. Methods The animal model was induced with oc- clusion of middle cerebral artery (MCAO) with suture embolus, and morroniside was then administered intragastrically at the dose of 30, 90 and 270 mg/kg for 7 d. Acetyl salicylic acid was taken as positive control drug. The content of Fib, and PT, APTT and TT were measured withrelated kits. Results Compared with the sham-operated group, the concentration of Fib significantly increased (P〈0.001) and PT, APTT and TT significantly shortened in the model group (1~〈0.001); however, compared with the model group, morroniside significantly decreased Fib content (P〈0.01) and prolonged PT, APTT and TT (P〈0.05). Conclusion Morroniside can antagonize the coagulation fi.mction in focal cerebral ischemia/reperfusion in rats.
