目的研究人参皂苷Rb1在厌氧和有氧条件下经肠道菌群作用下逐级脱糖基的代谢差异。方法利用UPLC-MS/MS研究人参皂苷Rb1在厌氧和有氧条件下经肠菌液的作用下,分别温孵0、2、4、8、12、24、48、72h后,人参皂苷Rb1及其脱糖基代谢物的变化情况。结果体外温孵实验中,人参皂苷Rb1在厌氧和有氧条件下经大鼠肠菌液作用下脱糖基生成人参皂苷Rd、F2、C-K,其中Rd→F2是Rb1在肠菌液的作用下脱糖基代谢的限速步骤。人参皂苷Rb1在大鼠肠道菌群作用下代谢途径为Rb1→Rd→F2→C-K。结论人参皂苷Rb1在有氧和厌氧条件下脱糖基代谢产物没有差异,有氧代谢速率快于厌氧代谢。
OBJECTIVE Research on the gradual deglycosylated metabolism of ginsenoside Rb1 by intestinal flora under aerobic and anaerobic conditions in vitro.METHODS The changes of Rb1 and its metabolites were detected by means of UPLCMS/MS incubated by intestinal bacteria under aerobic and anaerobic conditions in vitro after 0,2,4,8,12,24,48 hand 72hin vitro.RESULTS It was found that Rb1 was converted into three metabolites ginsenoside Rd,F2 and C-K by rat intestinal bacteria in vitro through gradual removal of glycosylation while Rd→F2 was the rate-limiting step.The metabolic pathway of Rb1 by intestinal bacteria was Rb1→Rd→F2→C-K.CONCLUSION The deglycosylated metabolites of Rb1 under aerobic and anaerobic conditions have no differences,but the metabolic rate under aerobic conditions is faster.
