目的 采用快速液相色谱LTQ-Orbitrap质谱联用技术对茯苓95%乙醇提取物中的化学成分进行分析.方法 色谱分离采用Thermo Hypersil Gold C18(3μm,150 mm×2.1 mm)色谱柱,以0.1%甲酸(A)-乙腈(B)梯度洗脱,质谱使用ESI离子源,负离子模式下采集数据.结果 通过一级和二级质谱分析并结合质谱碎裂规律及特征碎片信息,并通过比对对照品数据及相关文献,从茯苓95%乙醇提取物中初步分析了42个化学成分.结论 本方法分离度良好、灵敏度高,可用于茯苓中化学成分的定性分析,并有助于对茯苓进行进一步的活性成分研究和质量控制.
OBJECTIVE A method for qualitative analysis of triterpenes in the ethanol extracts of Poria cocos by UHPLCLTQ-Orbitrap mass spectrometry was established.METHODS Thermo Hypersil Gold C18 (3μm,150 mm×2.1 mm) column was applied in the UHPLC analysis of the triterpenes in Poria cocos,with acetonetrile-water (0.1% formic acid) as mobile phase.RESULTS Electrospray ionization (ESI)-LTQ-Orbitrap MS/MS detection was performed to obtain the MS spectra.Based on the high-resolution mass information,MS/MS fragmentation behaviors and chemical components from literatures,42 compounds were identified or tentatively characterized.CONCLUSION This method could be helpful for the quality control and further phytochemical studies of Poria cocos,providing a good example for the analysis of chemical constituents in traditional Chinese medicine.
