目的分析不同生长期当归中化学成分的动态变化。方法采用高效液相色谱法对当归样品中的化学成分进行分离分析。Agilent Zorbax SB-C18(4.6 mm×250 mm,5μm)色谱柱;流动相为水(A)-甲醇(B),洗脱梯度为0min,20%B;80 min,100%B;检测波长280 nm;体积流量0.8 mL/min;柱温30℃。结果不同生长期当归中阿魏酸、Z-藁本内酯、正丁烯基苯酞等成分的含有量变化明显,在第2年10~11月份时(传统采收期),其生物产量最大,阿魏酸含有量较高,Z-藁本内酯、正丁烯基苯酞含有量不高。结论当归中次生代谢产物的积累与生长周期密切相关。
AIM To analyse the dynamic accumulations of chemical constituents in Angelicae sinensis Radix from different cultured times.METHODS HPLC with diode array detector(HPLC-DAD) was performed on a Zorbax SB-C18 analytical column(4.6 mm×250 mm,5 μm).The mobile phase consisted of water(A) and methanol(B) in gradient elution mode(0 min,20% B,80 min,100% B) at the flow rate of 0.8 mL/min.The column temperature was set at 30 ℃.The detection wavelength was set at 280 nm.RESULTS The contents of ferulic acid,Z-ligustilide and n-butylidenephthalide in Angelicae sinensis Radix showed remarkable differences at different cultured times.The highest biomass yield was appeared in the roots harvested in October to November of the second year(the usual cultured times) accompanied with the higher ferulic acid content and with the lower Z-ligustilide and n-butylidenephthalide contents.CONCLUSION The accumulation of secondary metabolites in Angelica sinensis Radix is closely related to its growth cycle.
