克隆了肺炎克雷伯氏菌(Klebsiella pneumoniae)及大肠杆菌(Escherichia coli)各自的醛脱氢酶基因puuC和aldH,获得了重组肺炎克雷伯氏菌K.p(pET-PK-puuC)和K.p(pET-PK-aldH),这两株菌均能以甘油为唯一碳源生产3-羟基丙酸(3-HP).发酵实验结果显示K.p(pET-PK-puuC)的甘油转化率和单位菌体产量与野生菌相比分别提高了135%和18%;K.p(pET-PK-aldH)的3-HP产量达1.4.3 g/L,是K.p(pET-PK-puuC)的1.8倍,比野生菌提高了81%,甘油转化率和单位菌体产量比野生菌分别提高了150%和88%.
The aldehyde dehydrogenase coding genes puuC and aldH from Klebsiella pneumoniae DSM 2026 and Escherichia coli K-12 respectively have been cloned, and two recombinant strains K. p (pET-PK-puuC) and K. p (pET-PK-aldH) were acquired. Upon glycerol induction, both strains could produce 3-hydroxypropionie acid (3- HP) using glycerol as the sole carbon source. Compared with the wild type strain, the reeombinant strain K. p (pET-PK-puuC) showed increases of 135% and 18% in glycerol conversion ratio and yield on a biomass basis, re- speetively. The strain K. p(pET-PK-aldH) produced 1.43 g/L of 3-HP, which is 1.8 times that produced by K. p (pET-PK-puuC), and demonstrated increases of 81% , 150% and 88% in 3-HP production, glycerol conversion ratio and yield on a biomass basis, respeetively.
