为了解抗菌肽JH-3的抗菌谱、溶血性和生物信息,试验利用琼脂糖扩散法测定抗菌肽JH-3对不同细菌和真菌的抗菌活性,利用分光光度法检测抗菌肽JH-3对红细胞的溶解活性,用生物软件对抗菌肽JH-3进行生物信息学分析。结果表明:抗菌肽JH-3对大肠杆菌、鼠伤寒沙门菌、多杀性巴氏杆菌、副猪嗜血杆菌、胸膜肺炎放线杆菌、金黄色葡萄球菌、猪链球菌2型、白色念珠菌、青霉有抗菌活性,浓度小于4μg/mL时没有溶血性;二级结构为α螺旋,有3个磷酸化位点,没有糖基化和泛素化位点,有10种酶的酶切位点,为阳离子性疏水多肽。说明抗菌肽JH-3为无溶血性的螺旋型阳离子抗菌肽,抗菌谱广。
A study was conducted to research antibacterial spectrum, hemolysis and bioinformatics of antimicrobial pepfide JH - 3. Agarese diffusion method was used to determine antimicrobial activities against several strains of bacteria and fungi. Spectrophotometry method was used to determine hemolysis, and software was used to determine bioinformatics. The results showed that antimicrobial peptide JH -3 manifested anti- microbial activities against Escherichla coll, Salmonella typhimurium, Pasteurella multoeida, Hnemophilus parasuis, Actinabacllus pleuropneumoniae, Staphylococcus aureus, Streptococcus suis, Candida albicaus and Penicillium. In the condition of the concentration was less than 4μg/mL, antimicrobial peptide JH - 3 manifested non - hemolysis. Antimicrobial peptide JH - 3 presented α - helix as secondary structure, and three phosphorylation sites, except glycosytation and ubiqutin site. It manifested 10 kinds of enzyme cutting sites of cationic hydrophobic peptides. It was concluded that antimicrobial peptide JH - 3 was non - hemolytic and helical cation with a broad spectrum.
