目的探讨核因子KB(NF-kB)在星形细胞肿瘤组织中的表达及其与肿瘤细胞凋亡的关系。方法应用免疫组化方法检测85例星形细胞肿瘤组织及10例正常大脑组织中的NF-kB p65,并用Tunel法检测凋亡肿瘤细胞,分析NF-kBp65的表达与患者生存时间的关系。结果星形细胞肿瘤中NF—kB p65蛋白阳性表达率为60.0%,对照组中NF-kB p65无表达。NF-kB p65在低级别与高级别星形细胞肿瘤的表达相比,P〈0.05。随着肿瘤恶性程度的增加,肿瘤细胞凋亡指数(AI)增高,NF-kB p65与AI呈正相关关系(r=0.437,P〈0.01)。随访资料完整的45例中NF-kB p65阴性者3a生存率为72.2%,明显高于阳性者的33.3%(P〈0.05)。结论NF-kB是与星形细胞肿瘤相关的癌蛋白,NF-kB参与星形细胞肿瘤的凋亡。
Objective To evaluate the significance of nuclear factor kappaB (NF-kB) protein and its relationship to tumor apoptosis in astrocytic tumors. Methods The expressions of NF-kB p65 protein, apoptosis index (AI) in 85 cases of astrocytie, of which 45 cases with full materials were studied immunohistochemically, and 10 eases of normal brain tissue were served as the control group. Results None of the normal tissues exhibited NF-kB p65, In contrast, 51 cases astrocytie tumor specimens were positive for NF-kB p65 protein with a 60.0% positive rate. The expressions of NF-kB p65 protein and M were correlated to the differentiation ( P 〈 0. 05 ). The expression of NF-kB p65 was closely related with the expression of AI( r =0. 473, P 〈0.01 ). The 3 years survival rate of the positive group for NF-kB p65 protein was lower than that of negative group for NF-kB p65 protein. Conclusions NF-kB p65 is a new oncogenic protein related to the astrocytic tumors. It can be involved in tumor apoptosis and poor prognosis.
