目的研究镁离子(Mg^2+)对神经细胞兴奋性氨基酸N-甲基-D-天冬氨酸(NMDA)损伤的保护作用。方法采用原代培养的大鼠皮层神经细胞模型,给予NMDA损伤,加入不同浓度的Mg^2+,测定神经细胞损伤噻唑蓝(MTT)变化。结果Mg^2+对兴奋性氨基酸损伤保护作用在1.0~3.0mmol/L范围内存在,2.0mmol/L效果最可靠。结论在重型颅脑损伤的临床治疗中应尽早使用镁剂,剂量根据脑脊液浓度尽快调整到2.0mmol/L保护作用最好。
Objective To investigate the neuroproteetive effect of Mg^2+ for neurons damaged by NMDA. Methods By using primary cultured rat cortical neurons, the change in MTT of neurons damaged by NMDA and protective effect of Mg^2+ were observed. Results The neuroprotective effect of Mg^2 + related to the concentration of Mg^2+ between 1.0 - 3.0 mmol/L and effective time. 2.0mmol/L is the best concentration for neuroprotective effect. Conclusion Mg^2+ should be clinically applied as early as possible, and the dosage should be adjusted to 2.0 mmol/L based on its concentration in CSF.
