建立了反相高效液相色谱-内标法测定茶油中7种酚类物质的含量。以丁香酸为内标物,C18反相柱(4.6 mm×250 mm,5μm)为分析柱,甲醇-水(0.2%)为流动相,流速1.0 mL/min;检测波长280 nm,柱温25℃。7种酚类物质在各自的线性范围内具有良好的线性关系,相关系数为0.998~0.999,检出限为0.008~0.028μg/g。7种酚类在茶油中的平均回收率为76.783%~88.956%。结果表明,在选定的7中酚类物质中茶油只含有其中的4种,分别是没食子酸、儿茶素、4-羟基苯乙酸、槲皮素,而且这4种物质在8个油样中的含量各异,这可能与茶油的加工方法扣品种有关。该方法准确,适用性好,可用于荼油中酚类物质的定性与定量分析。
The determination method for content of phenol compounds in camellia oil has been established by reverse phase high performance liquid chromatography ( RP - HPLC ) adopting syringic acid as internal standard. The RP- HPLC was conducted on C18 reverse phase column (24.6 mm ×250 mm,5 μm) at 25 ℃ with methanol -water (0.2%) as mobile phase. The flow rate of mobile phase was 1.0 mL/min. The detective wave- length was set up as 280 nm and column temperature at 25 ℃. It was proved that seven selected phenolic compounds had good linear relationships to the concentration range with correlation coefficients of ranging from 0. 998 to 0.999. The detection limits distributed from 0. 008 to 0. 028μg/g. The average recoveries of seven phenols in camellia oil stayed between 83. 023% and 91. 750%. The results indicated that four kinds of phenols have been found in tasted camellia oil sampies, including gallic acid, catechins, 4 - hydroxyphenylaeetic acid and quercetin with varied contents due to different processing methods. The content of phenols which existed in 8 camellia oil sample was different; it probably attributed to the difference of processing methods or varieties of camellia oils. These factors suggested that the above method with relative high accuracy and sensitivity could be applied in qualitative and quantitative analysis of camellia oil.
