TRIM5α(tripartite motif protein5-alpha,TRIM5α)是一种能够直接识别病毒衣壳的细胞质蛋白质,它通过影响入侵病毒粒子衣壳的脱落干扰病毒的有序脱壳,从而抑制HIV-1感染后的逆转录过程。各种区域突变型TRIM5α对抑制HIV-1逆转录具有的不同作用,启发了利用人内源性TRIM5α对AIDS进行治疗的思路。本文综述了TRIM5α抗HIV-1感染的功能结构域、作用机制及相关的基因治疗策略。
Tripartite motif protein 5-alpha (TRIM5α) is a cytoplasmic protein that directly recognizes HIV capsid and disrupt the ordered process of viral uncoating, eventually, interfering HIV-1 reverse transcription and further replication. The mutants of TRIM5α play different roles in the inhibition of HIV-1 reverse transcription, which inspires people to treat AIDS by using human endogenous TRIM5α. The interaction domains, action mechanism and related gene therapy strategy of TRIM5α to inhibit HIV-1 infection were reviewed in this paper.
