目的观察小鼠骨折愈合过程中FGFRs的表达变化。方法按标准闭合性骨折制作模型的方法来制作骨折模型,分别从5个时间点(骨折后第3天、7天、14天、21天和28天)观察骨痂组织中FGFRs mRNA的表达情况。结果 FGFR1骨折后3天开始显著升高,14天达到峰值并持续到骨折后28天(P〈0.001);FGFR2骨折后7天显著升高(P〈0.01),FGFR3在骨折后7天达到峰值后逐渐恢复正常。结论在骨折愈合过程的不同阶段,FGFRs存在差异表达,可能在成年骨折愈合中发挥重要作用。
Objective To investigate the expressions of FGFRs mRNAs in mice fracture healing.Methods Closed proximal tibia fracture was created in 2-month-old mice.Callus tissue properties was analyzed at 1,2,3 weeks post-fracture histology.RNA was isolated from callus tissues,and the expression levels of bone formation-related genes were evaluated by real time PCR.Results FGFR1mRNA levels upregulated on day 3 and reached maximum expression on day 14,then began to decline,but remained at relatively high levels even on day 28.FGFR2 mRNA levels upregulated on day 7.FGFR3 mRNA levels reached the peak on day 7 then declined to normal on day 28.Conclusion These results indicate the expressions of FGFRs at different stage of the healing process are different,which may participate in the regulation of fracture healing.
