目的探讨高血压大鼠肾损伤时调节性T淋巴细胞(regulatory T lymphocytes,Treg)及连接蛋白43(connexin 43,Cx43)在外周血的表达变化。方法选取16~17周龄雄性自发性高血压大鼠(spontaneous hypertension rats,SHR)和正常Wistar京都(Wistar-Kyoto,WKY)大鼠,各17只,测量血压后,采用苏木素-伊红(hematoxylin and eosin,HE)染色观察大鼠肾脏组织的病理学改变,应用流式细胞术检测大鼠外周血中CD4+CD25+调节性T淋巴细胞以及Cx43在CD4+和CD8+T细胞的表达比率。结果与WKY大鼠比较,SHR肾脏血管壁增厚,部分血管周围炎性细胞浸润和肾小球萎缩等肾损伤现象;WKY大鼠和SHR外周血中CD4+CD25+调节性T淋巴细胞所占CD4+T细胞的频率分别为(3.1±0.1)%和(1.9±0.1)%,两者间有显著性差异(P〈0.01);WKY大鼠和SHR外周血中CD4+Cx43+的表达比率分别为(0.76±0.20)%和(1.83±0.34)%;CD8+Cx43+的表达比率分别为(25.33±1.71)%和(36.46±2.87)%,差异均有统计学意义(P〈0.05)。结论自发性高血压大鼠肾损伤伴随其外周血中CD4+CD25+调节性T淋巴细胞的频率下调,且Cx43在CD4+和CD8+T淋巴细胞的表达增加。
The study was designed to investigate the different expression of regulatory T lymphocytes(Treg)and connexin 43(Cx43) in peripheral blood of hypertension rats with renal injury. Sixteen to seventeen-week-old male spontaneous hypertension rats(SHR) and normal Wistar-Kyoto(WKY) rats(n=17, respectively) were recruited in this study. After blood pressure measurement, hypertension-induced renal pathological changes were analyzed by hematoxylin and eosin(HE) staining; CD4+CD25+Treg cell populations and the expression of Cx43 in CD4+and CD8+lymphocyte of peripheral blood from WKY rats and SHRs were detected by flow cytometry. HE staining showed that there were significant thickening of vascular wall, inflammatory cell infiltration into part of the blood vessels and glomerular atrophy in SHRs, but not in WKY rats. The frequency of CD4+CD25+Treg cell in CD4+cells of peripheral blood from WKY rats and SHRs were(3.1±0.1)% and(1.9±0.1)%, which were statistical difference(P〈0.01). Most importantly, flow cytometry indicated the radio of Cx43 expression in CD4+lymphocyte of peripheral blood from WKY rats and SHRs were(0.76 ±0.20)% and(1.83 ±0.34)%, while in CD8+lymphocyte were(25.33 ±1.71)% and(36.46±2.87)%, which were statistical significance(P〈0.05). Taken together, along with renal injury in SHR, the frequency of CD4+CD25+Treg cell would decreased and the expression of Cx43 in CD4+or CD8+lymphocyte would increased in peripheral blood.
