目的:建立以高效液相色谱法测定广西产美味猕猴桃根中三萜化合物含量的方法。方法:以2a,3a,24-三羟基-12-烯-28-乌苏酸为标准品,色谱柱为Thermo Hypersil BDSC18(250mm×4.6mm,5um),柱温为20℃,流动相为甲醇-0.2%磷酸水溶液(73:27),流速为1.0mL·min^-1,检测波长为210nm。结果:2α,3α,24-三羟基-12-烯-28-乌苏酸的检测浓度在0.025-0.200mg·mL^-1范围内与峰面积积分值呈良好的线性关系(r=0.9998);平均回收率为99.79%,RSD=1.54%(n=6)。结论:本方法准确、可靠、重复性好,可用于广西产美味猕猴桃根药材的质量控制。
OBJECTIVE: To establish an HPLC method for the determination of triterpenoid in the roots of.4ctinidia deliciosa from Guangxi. METHODS: With the content of 2a,3a,24-trihydroxyursa-12-en-28-oic acid used as index. The separation was performed on Thermo Hypersil BDS C18 column (250 mm×4.6 mm, 5 um) with 2a,3a,24-trihydroxyursa-12-en-28-oic acid as standard substance. The mobile phase consisted of methanol-0.2% phosphoric acid (73:27) with column temperature set at 20 ℃. The flow rate was set at 1.0 mL.min^-1 and detection wavelength was 210 nm. RESULTS: The linear range of 2a,3ct,24-trihydroxyursa-12-en-28-oic acid were 0.025-0.200 mg. mL^-1 (r=0.999 8). The average recovery was 99.79% (RSD=1.54% ,n=6). CONCLUSION: The method is accurate, reliable and reproducible for the quality control of the roots of A. deliciosa from Guangxi.
