本试验旨在研究共轭亚油酸( CLA)和α-亚麻酸( ALA)对于HepG2细胞核转录因子NF?E2相关因子( Nrf2)及其调控的谷胱甘肽硫转移酶A1( GSTA1) mRNA 和蛋白质表达的影响。试验组采用浓度为0.2、0.5和1.0 mmol/L的CLA和ALA作用HepG2细胞24 h,采用荧光定量PCR法测定mRNA的表达量,蛋白质印迹法测定蛋白质的表达量。结果表明,CLA浓度分别为0.2和0.5 mmol/L 时,Nrf2和 GSTA1 mRNA 和蛋白质表达量与对照组相比极显著增加( P<0.01),并随CLA浓度增加呈下降趋势;采用ALA作用的细胞,Nrf2和GSTA1 mRNA和蛋白质表达量与对照组相比,随ALA浓度的增加呈现极显著上升( P<0.01)。由此可见,CLA在较低浓度时,诱导Nrf2和GSTA1 mRNA和蛋白质表达量作用较强,而随ALA浓度增加Nrf2和GSTA1 mRNA和蛋白质的表达量逐渐增多。
The purpose of the study was to explore the effects of conjugated linoleic acid ( CLA) and α-linole-nic acid ( ALA) on mRNA and protein expression of nuclear transcription factor nuclear factor erythroid 2-relat-ed factor 2 (Nrf2) and glutathione S-transferase A1 (GSTA1) in HepG2 cells. The experiment groups were used 0.2, 0.5 and 1.0 mmol/L CLA and ALA treated HepG2 cells for 24 h, the mRNA expression levels were quantified by quantitative reverse transcription PCR analysis, and the protein expression levels were detected by western blotting method. The results showed as follows:compared with the control group, the mRNA and pro-tein expressions of Nrf2 and GSTA1 were significantly incresed in cells treated with CLA with the concentration of 0.2 and 0.5 mmol/L ( P〈0.01) , and with the CLA concentration increased, the mRNA and protein expres-sions of Nrf2 and GSTA1 were decreased. However, when HepG2 cells were treated with ALA, the mRNA and protein expressions of Nrf2 and GSTA1 were significantly increased with the ALA concentration increased of compared to control group ( P〈0.01) . In conclusion, CLA with a relatively lower concentration elevates the mRNA and protein expressions of Nrf2 and GSTA1, on the contrary, the mRNA and protein expressions of Nrf2 and GSTA1 are increase gradually with the CLA concentration increased.
