本文旨在建立一种同时检测小鼠血液和肝脏组织中脂肪酸含量的气相色谱方法。以Floch法萃取肝脏中的脂肪酸,13%BF3-甲醇溶液进行脂肪酸的甲酯化,采用DB-23色谱柱和FID检测器进行检测,程序升温梯度分离。17种脂肪酸线性关系良好;小鼠血清和肝脏平均回收率在82.65%-100.72%和80.72%-100.62%,精密度在0.53%-5.51%和0.58%-5.74%。脂肪酸的检测限在100-300 ng/mL,定量限在150-450 ng/mL。
To establish a gas chromatography(GC)method for the determination of fatty acids in both serum and liver tissues.Folch method was adopted to extract lipids in liver,fatty acids were methylesterized by 13% BF3-methanol solution,then separat-ed and measured by GC system equipped with a DB-23 glass capillary and a FID. A temperature program was designed. Therewere good linear relationships. The average recovery rates of the method were 82.65% to 100.72% in serum and 80.72% to100.62% in liver.The precisions ranged from 0.53% to 5.14% in serum and 0.58% to 5.70% in liver. The limit of detection rangewas 100 ng/mL to 300 ng/mL,and the limit of quantity range was 150 ng/mL to 450 ng/mL.
