高脂饲料对小鼠血清脂肪细胞因子瘦素和脂联素的影响

本研究采用高脂饲料诱导小鼠,建立非酒精性脂肪肝模型并分析其对脂肪细胞因子瘦素和脂联素的影响。雄性IcR小鼠12只,随机分成对照组和高脂组,每组6只,分别饲喂基础饲料和高脂饲料,于连续喂养4周和8周后,眼球采血处死,称取小鼠的体重和肝重,计算肝指数,并测定血清中的生化指标。结果显示:与对照组相比.高脂组小鼠的肝指数极显著升高(P〈0.01),高脂组的谷丙转氨酶(ALT)、谷草转氨酶(AST)、胆固醇(TC)、葡萄糖(GLu)、碱性磷酸酶(ALP)、瘦素和胰岛素水平极显著或显著高于对照组(P〈0.01或P〈0.05),而甘油三酯的变化不明显(P〉O.05),脂联素的水平极显著低于对照组(P〈0.01)。试验结果表明,高脂饲料饲喂小鼠会引起模型组小鼠发生脂肪肝变化.并产生胰岛素抵抗、瘦素抵抗和低脂联素血症。

A non-alcoholic steatohepatitis animal model induced by high-fat diet was established,and its effects on leptin and adiponectin were studied.12 male ICR mice were randomly divided into 2 groups,each group contained 6 mice.The control group was fed with standard diet and the model group fed with high-fat diet.The blood samples of two groups were collected by removal of the mouse eye after 4 weeks and 8 weeks feeding.Then the body and liver were weighted.The serum were assayed at the 4th and 8th week by automatic biochemical analyzer.Serum leptin,adiponectin and insulin levels were measured by enzyme-linked immunosorbent assay (ELISA).The results showed that:compared with control group ,the level of liver index significantly increased.The levels of alanine aminotransferase (ALT),aspartate aminotransferase (AST), total cholesterol (TC), glucose (GLU), alkaline phosphatase (ALP), leptin and isulin in serum were significantly higher than those of control group(P 〈 0.01 or P 〈 0.05 ).Triglyceride(TG) did not change significantly (P 〉 0.05).Serum adiponectin level of high-fat group was significantly lower than that of control group (P 〈 0.01).So feeding with high-fat diet could induce steatoheoatitis in mouse rapidly and develop insulin resistance,leptin resistance and hypoadiponectinemia.
