目的:通过观察艾灸对Hp胃黏膜炎性损伤大鼠胃黏膜损伤指数和胃组织匀浆液中炎性细胞因子TNF-α、IL-1β、IL-10、IL-12含量的影响,初步揭示艾灸干预Hp胃黏膜炎性损伤,保护胃黏膜损伤的机制。方法:50只健康SD大鼠随机分为5组,即空白组(A组)、Hp胃炎模型组(B组)、模型+艾灸穴位组(C组)、模型+艾灸非穴点组(D组)和模型+电针穴位组(E组)。采用Hp灌胃造模。取大鼠足三里、中脘、关元、脾俞和胃俞穴进行艾灸。参照GUTH法计算胃黏膜损伤指数(UI),采用酶联免疫法检测大鼠胃组织匀浆液中TNF-α、IL-1β、IL-12、IL-10含量。结果:艾灸可降低Hp胃黏膜炎性损伤大鼠胃黏膜损伤指数(P〈0.01),可降低胃组织匀浆液TNF-α、IL-1β、IL-12含量(P〈0.05或P〈0.01),增高IL-10含量(P〈0.01),与模型组、艾灸对照点组和电针组比较有显著性差异。结论:通过抑制胃黏膜局部组织TNF-α、IL-1β、IL-12释放,促进IL-10释放,从而减轻胃黏膜炎症反应,可能是艾灸降低Hp胃炎大鼠胃黏膜炎性损伤的机制之一。
Objective:To explore the effect of the content of the UI and the content of TNF-α,IL-1β,IL-12,IL-10 gastric tissue homogenate of Hp gastritis in rats with moxibustion treatment,to reveal the mechanisms of intervention Hp gastric inflammatory injury and protect the gastric mucosa in rats with moxibustion treatment.Methods:50 healthy rats were randomly assigned into 5 groups,namely control group(group A),Hp model group(group B),moxibustion at acupoints group(group C),moxibustion at non-acupoints(group D) and electro-acupuncture acupoints group(group E).The gastritis model was made with Hp Gavage.To moxibustion at Zusanli,Zhongwan,Guanyuan,Pishu,Weishu in rats,the ulcer index(UI) was evaluated by Guth method,the content of TNF-α,IL1β,IL-12,IL-10 of gastric tissue homogenate were detected by enzyme-linked immunosorbent assay.Results:After moxibustion treatment,the UI were significantly decreased(P0.01),the content of TNF-α,IL-1β,IL-12 gastric tissue homogenate were significantly decreased(P0.05 or P0.01),the content of gastric tissue homogenate IL-10 were significantly increased(P0.01) compared with Hp model group,moxibustion at non-acupoints and electro-acupuncture acupoints group.Conclusion:Moxibustion could increase the content of IL-10 and decrease the content of TNF-α,IL-1β,IL-12 in gastric tissue homogenate,which suggested moxibustion could adjust the release of proinflammatory cytokine and suppression of inflammatory cytokines,intervene the mucosal inflammatory injury of rats with Hp gastric,that may be the one of mechanisms of intervention Hp gastric inflammatory injury and protect the gastric mucosa in rats with moxibustion.
