目的初步揭示艾灸干预幽门螺旋杆菌(Hp)胃黏膜炎性损伤,保护胃黏膜损伤的机制。方法50只健康SD大鼠随机分为5组,即空白组、Hp模型组、艾灸组、艾灸对照点组和电针组。采用Hp灌胃造模。采用酶联免疫法检测大鼠胃组织匀浆液中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-12(IL-12)、白细胞介素-10(IL-10)含量。结果艾灸预处理可降低Hp胃黏膜炎性损伤大鼠胃黏膜损伤指数(P〈0.01),可降低胃组织匀浆液炎性细胞因子TNF-α、IL-1β、IL-12含量(P〈0.05或P〈0.01),增高胃组织匀浆液炎性细胞因子IL-10的含量(P〈0.01),与Hp模型组、艾灸对照点组和电针组比较有显著性差异。结论通过抑制胃黏膜局部组织促炎因子TNF-α、IL-1β、IL-12释放,促进抑炎因子IL-10释放,从而减轻胃黏膜炎症反应,保护胃黏膜损伤的作用,可能是艾灸预处理降低Hp胃黏膜炎性损伤大鼠胃黏膜炎性损伤的机制之一。
Objective To explore the mechanism of moxibustion in intervening inflammatory injury of Helicobacter pylori (lip) gastritis and protecting gastric mucosa. Methods Health SD rats ( n = 50) were randomly divided into blank group, Hp model group, acupoint moxibustion group, non-acupoint moxibustion group and electroacupuncture group (each n = 10 ). The rat model of lip gastritis was established by intragastrically giving of Hp. The content of tumor necrosis factor-or (TNF-α), interleukin-1 β (IL-1 β) , interleukin-12 (IL-12) and interleukin-10 (IL-IO) in gastric homogenate were detected by applying enzyme-linked immunosorbent assay (ELISA). Results The pretreatment of moxibustion reduced gastric mucosal injury index (P 〈 0.01 ), decreased the content of TNF-α, IL-1 β and IL-12 (P 〈0.05 or P 〈0.01 ), and increased the content of IL-10 (P 〈0.01 ) compared with Hp model group, acupoint moxibustion group and electroacupuncture group. Conclusion The pretreatment of moxibustion can relieve the gastric mucosal injury of Hp gastritis, and the mechanism may be related to inhibiting the release of pro-inflammatory cytokines (TNF-α, IL-1β and IL-12) and improving therelease of anti-inflammatory cytokines (IL-10) for relieving the inflammatory reaction and protecting gastric mucosa.
