[摘要]目的观察艾灸对幽门螺杆菌胃炎模型大鼠胃黏膜组织形态学变化、血清IL-6、IL-8、TNF-α、Hp—IgG、CD3^+、CD4^+、CD8^+的影响,探讨艾灸干预HP胃炎胃粘膜损伤的机制。方法40只SD大鼠随机分为4组,每组10只,即A组(空白组)、B组(Hp胃炎模型组)、C组(艾灸+模型组)、D组(电针+模型组)。除A组外其余各组大鼠均以NaHCO:消炎痛+Hp灌胃造模。各组经相应的干预后,HE染色光镜下观察大鼠胃黏膜组织形态学变化,Elisa法测定血清中IL-6、IL-8、TNF—α、Hp—IgG的含量,流式细胞术测CD3^+、CD4^+、CD8^+、CD4^+,CD8^+值。结果与A组相比,B组大鼠血清IL-6、IL-8、TNF—α、Hp—IgG、CD8^+明显增高(P〈0.01),CD3^+、CD4^+、CD4^+/CD8^+明显降低(P〈0.01)。与B组相比,C组血清IL-8、TNF-α、IL-6含量、Hp—IgG、CD8^+明显降低(P〈0.05或0.01),CD3^+、CD4^+、CD4^+/CD8^+明显升高(P〈0.05),D组各指标无明显变化(P〉0.05)。与D组相比。C组IL-6、IL-8、Hp—IgG明显降低(P〈0.05),CD4^+/CD8^+明显升高(P〈0.05)。结论艾灸防治幽门螺杆菌胃炎的作用可能通过两种途径实现。抑制促炎细胞因子IL-6、IL-8、TNF-α的分泌,促进免疫因子CD3^+、CD4^+产生,抑制CD8^+分泌,且艾灸对胃黏膜组织保护作用比电针更明显。
Objective To explore the effects of moxibustion on the morphologic changes of gastric mucosa tissues and expression of serum IL-6, IL-8, TNF-α, Hp-IgG, CD3^+, CD4^+, CD8^+, CD4^+/CD8^+in Helicobacter pylori(Hp)-associated gastritis model rats,to reveal the mechanisms underlying the protective effect of moxibustion against gastric inflammatory injury. Methods 40 healthy SD rats were randomly divided into 4 groups:the group A(blank group), group B(Hp model group), group C(moxibustion model groups) and group D(electro-acupuncture model group), 10 rats in each group.The rats except for the group A were modeled by oral gavage with Na HCO3+indomethacin+Hp. After the intervention,the morphologic changes of gastric mucosa tissues in rats were observed by HE staining. The contents of serum IL-6, IL-8,TNF-α, Hp-IgG were detected by ELISA, and Contents of CD3^+, CD4^+, CD8^+and CD4^+/CD8^+were detected by flow cytometry. Results Compared with group A, the expression of serum IL-6, IL-8, TNF-α, Hp-IgG, CD8^+in group B were significantly increased(P〈0.01), and contents of CD3^+, CD4^+and CD4^+/CD8^+were decreased obviously(P〈0.01). Compared with group B, the expression of serum IL-6, IL-8, TNF-α, Hp-IgG, CD8^+in group C were significantly decreased(P〈0.05 or0.01), contents of CD3^+(P〈0.05)、CD4^+(P〈0.05) and CD4^+/CD8^+were increased significantly(P〈0.05), no significant difference in group D(P〈0.05). Compared with group D, the expression of IL-6, IL-8, Hp-IgG in group C were significantly decreased(P〈0.05 or 0.01), CD4^+/CD8^+increased significantly(P〈0.05). Conclusion The mechanisms of moxibustion in treatment of H. p ylori-induced gastric mucosal inflammatory injury were probably through two pathways, Inhibition of proinflammatory cytokine IL-6, IL-8, TNF-αsecretion, and promoting immune factor CD3^+、CD4^+secretion to inhibit the secretion of CD8^+.
