目的测定比格犬血浆中低浓度的盐酸二甲双胍,研究盐酸二甲双胍在消化道各部位的吸收情况。方法采用DikmaDiamonsilC18柱(150mm×4.6mm,5μm),流动相为乙腈-10.0mmol·L-1磷酸二氢钠水溶液(含7.5mmol·L。十二烷基磺酸钠,pH5.5)(32:68),柱温30℃,检测波长235nm,流速1.0ml·min-1,内标法检测。结果盐酸二甲双胍与内标分离良好,线性范围0.05~4.0μg·ml-1;在比格犬的胃、小肠和结肠3个部位释药得到的药物动力学参数差异明显。结论所建方法灵敏、准确度好,测定快速、简便。
OBJECTIVE To develop a method for the quantification of low concentration metformin hydrochloride in Beagle dog's plasma by HPLC. METHODS HPLC method was conducted on a Dikma Diamonsil C18 (150 mm ×4.6 mm,5 μm) with acetonitrile - 10.0 mmol· L-1 sodium dihydrogen phosphate ( 7.5 mmol· L - 1 SDS, pH5.5 ) ( 32 : 68 ) as a mobile phase with column temperature at 30℃, detection wavelength at 235 nm and flow rate of 1.0 ml. min-l. The internal standard method was used. RESULTS The identified characteristics of Mefformin hydrochloride and the internal standard were distinct. The linear range was 0.05 - 4.0 μg· ml-1. The absolute recovery rates were within 90.93% - 97.45% and the relative recovery rates were within 99.78% - 107.90%. The intra RSD was less than 3.85% and the inter - precision was less than 5.63%. The pharmacokinetic parameters of Mefformin hydrochloride in the Beagle dogs stomach, small intestine and colon were significantly different. CONCLUSION The method is sensitive~ accurate, rapid and simple, which can be adopted to determine the low concentration of mefformin hydrochloride in Beagle dog's plasma and for its pharmacokinetic study.
