目的:探讨三硝基苯磺酸(TNBS)所致狭窄对大鼠结肠运动模式的影响。方法:雄性SD大鼠32只随机分为TNBS造模组、TNBS+思密达组、思密达组及正常对照组,各组大鼠分别给予2%TNBS灌肠、2%TNBS灌肠+思密达灌胃、思密达灌胃及正常饲养。8d后处死并用时空图记录各组大鼠离体结肠的运动模式。结果:TNBS组远端结肠出现明显狭窄,长距离收缩运动(LDC)无法下传至远端结肠,狭窄部远端结肠出现独立于近端结肠运动模式的节律性运动模式,提示全结肠运动协调性丧失;狭窄部位以上近端结肠观察到LDC、节律性运动复合波(RPMCs)、微小波和分节运动等模式;TNBS+思密达组远端结肠未出现明显狭窄,全结肠协调性完整。结论:TNBS可致结肠神经源性损害、影响全结肠协调性,思密达可防止TNBS造成的结肠狭窄发挥神经保护作用,维护全结肠协调性。
Objective:To explore the effects of TNBS-induced stricture on colonic motor patterns.Methods:Thirty-two SD rats were divided randomly and equally into four groups:A,B,C,and D according to the treatment TNBS,TNBS+Smecta,Smecta,and normal saline(normal control),respectively.All rats were killed 8days later and using spatiotemporal mapping to investigate the motor activities of the proximal colon.Results:Formation of strictures could be seen in the model of group A and no long distance contraction(LDC)develops.There were more complexes coming from the stricture,which were independent of the proximal colon;Motor patterns like rhythmic propulsive motor complexes,ripples,segmentation,proximal contractions and LDCs were observed in proximal colon;No obvious stricture appeared in group B and no changes in colonic motor patterns were found.Conclusion:The TNBS-induced stricture affects colonic motor patterns,but dioctahedral smectite can obviously prevent the TNBS-induced stricture,protectneurogenic components and maintain the coordination of movement of whole colon.
