目的:细胞黏附性能是评价神经组织工程基质材料的重要指标,拟对包含神经活性多肽片断的IKVAV多肽两亲性分子在体外进行自组装,并且检测其对PC12细胞黏附的影响。 方法:实验于2005-12/2006—03在华中科技大学附属协和医院骨科实验室完成。IKVAV多肽两亲性分子委托上海波泰生物科技公司合成,并用高效液相色谱仪和质谱仪进行纯化和分析。在IKVAV多肽两亲性分子中加入稀盐酸降低pH值引发其自组装,用不同浓度的IKVAV自组装材料包被培养板,培养PC12细胞,检测IKVAV对PC12细胞黏附的影响结果:①当IKVAV自组装材料的密度为0.58-15.0μg/cm^2 。时,能明显增强PC12细胞黏附,而且在一定的范围内随密度增加PC12细胞的黏附率升高。②在1h和3h各IKVAV密度组的细胞黏附率之间差异无显著性: 结论:IKVAV能够促进PC12细胞黏附,而且有一定的量效关系。同时,IKVAV促进PC12细胞黏附的作用迅速、稳定。

英文摘要:

AIM: The capability of cell adherence is an important indicator in evaluating the scaffolds of neural tissue engineering. To conduct in vitro self-assembly of neural bioactive polypeptide isoleucine-lysine-valinealanine-valine (IKVAV), and detect its effect on attachment of PC12 cells. METHODS: The experiment was conducted in the Laboratory of Orthopaedics, Union Hospital Affiliated to Tongji Medical College, Huazhong University of Science and Technology from December 2005 to March 2006. IKVAV-PA was synthesized by Shanghai Botai Biological & Technical Corporation, which was then purified and tested with high-performance liquid chromatography (HPLC) and mass spectrometry (MS) respectively. The diluted hydrochloric acid was added into IKVAV-PA solution to reduce the pH value and accordingly induce self-assembly. Cell culture plates were coated with different concentrations of IKVAV self-assembly peptide. PC 12 cells were cultured and added to IKVAV coated plates. The effect of IKVAV on the attachment of PC 12 cells were tested. RESULTS: ①IKVAV-PA could significantly promote the attachment of PC12 at the density ranged from 0.58 μg/cm^2 to 15.0 μg/cm^2. Moreover, within a certain range, the attachment rate of PC12 cells increased with the density increased. ②There was no significant differences in the attachment rate at the 1^st and 3^rd hour among each IKVAV groups. CONCLUSION: IKVAV can promote the attachment of PC12 cells, which is in a dose-effect relationship. Meanwhile, the effects of IKVAV on the attachment of PC12 cells is rapid and stable.
