目的研究成神经细胞分化不同阶段的骨髓间充质干细胞(BMSCs)向干细胞因子(SCF)的趋化性迁移。方法 Percoll密度梯度离心法分离培养SD大鼠BMSCs,体外传代并扩增纯化,通过免疫荧光染色及多向分化对其进行鉴定。采用碱性成纤维生长因子(bFGF)、丁羟基茴香醚(BHA)联合诱导剂对BMSCs进行成神经细胞诱导,观察分化各时期的细胞形态变化。利用Dunn chamber研究SCF对不同分化阶段BMSCs趋化性迁移的影响。结果 Dunn chamber迁移实验显示,SCF诱导实验组细胞的迁移速率和迁移效率明显高于对照组(P〈0.05),表明SCF对BMSCs具有趋化作用。此外,分化不同状态的BMSCs向SCF的趋化迁移程度也不同。结论 BMSCs可以分化为神经样细胞,其对SCF的趋化性迁移与分化状态密切相关。
Objective To study the effect of stem cell factor(SCF) on the migration of bone marrow mesenchymal stem cells(BMSCs) in neuroblast differentiation.Methods BMSCs were isolated from the bone marrow of SD rats by Percoll gradient centrifugation.bFGF and BHA were used to induce BMSCs to differentiate into neural-like cells.The morphology of BMSCs during neuroblast differentiation was observed with inverted microscope.Cell migration was analyzed by time-lapse video analysis using Dunn chamber.Results Dunn chamber analysis it was revealed that both the migration speed and efficiency of cells induced by SCF were signifficantly higher than the controls(P〈0. 05),demonstrating the chemotactic effect of SCF on BMSCs.BMSCs at different differentiation states showed different tropisms for SCF.Conclusion BMSCs can be induced to be differentiated into neuron-like cells,Tropism of BMSCs for SCF was closely correlated to their differentiation states.
