本实验旨在探究Nkx2—5基因在小鼠黑色素细胞中是否表达,同时通过生物信息学的方法预测其是否与调控黑色素生成的基因有相互作用。结果表明:转录因子Nkx2—5在小鼠黑色素细胞中表达,其编码的蛋白作为转录因子与下游基因作用的主要位点在144~196位氨基酸残基处,且其在黑色素生成中的关键基因MITF、TYR、TYRP1、TYRP2的启动子中都有作用位点,同时Nkx2—5蛋白与对神经嵴分化有重要影响的Hand1、Hand2存在相互作用,故可以推测转录因子Nkx2—5在黑色素细胞的黑色素生成中发挥着重要的作用,进而为从分子水平解释动物被毛白化的发病机理提供新的思路。
This assay aimed to study whether the Nkx2-5 expressed in the mouse melan ocytes and, through the bioinformatics, predict whether the Nkx2-5 can influence the production of melanin. The results showed the Nkx2-5 gene expressed in mouse melanocytes, and the target gene binding sites of Nkx2-5 were mainly distributed in 144- 196 amino acids, there were binding sites of Nkx2-5 at MITF, TYR, TYRP1, TYRP2 promoter, meanwhile, Nkx2-5 could interact with Handl, Hand2 which were important to the differentiation of neural crest. So we can infer the transcription factor of Nkx2-5 has important influence in the production of melanin, furthermore, this can provide a new idea to explore the pathogeny of albinism.
