目的:探讨糖肾方对糖尿病肾病大鼠纤溶功能的影响。方法:Wistar大鼠利用单侧肾切除合并腹腔注射链脲佐菌素诱导大鼠糖尿病肾病模型,随机分为模型组、蒙诺组(0.833g/kg)、糖肾方大剂量组(2.67g/kg)和糖肾方小剂量组(1.33g/kg),每组10只;另取10只进行假手术后腹腔注射柠檬酸缓冲液作为空白组。各实验组每日灌胃给药1次,连续给药12周。每4周检测血糖和尿蛋白/肌酐水平。实验结束时处死大鼠,取血检测总胆固醇、甘油三酯、组织型纤溶酶原激活物(TPA)、纤溶酶原活化物抑制因子(PAI)、纤维蛋白降解产物(FDP)水平,取肾组织进行病理组织学观察。结果:糖肾方大剂量能显著降低模型大鼠尿蛋白/肌酐(P〈0.05),减少血清胆固醇和甘油三酯水平(P〈0.05,P〈0.01);糖肾方大、小剂量均可显著上调TPA含量,下调PAI及FDP水平;减轻模型大鼠肾小球和肾小管间质损害。结论:糖肾方对糖尿病肾病大鼠具有肾保护作用,其作用机制可能与降低血脂代谢,调节纤溶功能有关。
Objective: To investigate the effect of Tangshen decoction therapy on fibrinolysis function in diabetic nephropathy rats. Methods: To establish diabetic nephropathy model rats, and rats were randomly divided into model group, Monopril group (0.833g/kg), high dosage of Tangshen decoction group (2.67g/kg) and low dosage of Tangshen decoction group (1.33g/kg) with 10 rats in each group. Another 10 rats were sham operated as normal control group. Rats were orally administered once a day for 20 weeks. Blood glucose and albumin/creatine levels were measured. At the end of the experiment, blood samples were taken from abdominal aorta for the detection of indices of total cholesterol (TC), triglyceride (TG), TPA, PAI, FDP etc. Kidney samples were taken for the observation of pathology. Results: High dosage of Tangshen decoction group significantly decreased albumin/creatine (P0.05) , TC (P0.05) and TG (P0.01) levels in the serum, regulated T-PA, PAI, FDP levels in the serum, effectively reduced the glomerlosclerosis index (P0.01) and the renal interstitial macrophage (P0.01) . Conclusion: Tangshen decoction has the good rapeutic effect on diabetic nephropathy rats, the main mechanisms of which are related to improving lipid metabolism and regulating fibrinolysis function.
