目的:探讨糖肾方对自发性2型糖尿病模型OLETF大鼠肾脏的保护作用以及对血中白细胞介素-10、肿瘤坏死因子-α和多巴胺的影响。方法:OLETF大鼠随机分为模型组、糖肾方组(1.6g/kg)、蒙诺组(1.0mg/kg)、LETO大鼠为正常对照组。各组动物从12周龄起进行灌胃给药,连续给药44周。实验结束,大鼠56周龄时,测定血糖和24h尿蛋白水平、血清中白细胞介素-10、肿瘤坏死因子-α、多巴胺。取肾组织观察肾组织形态学改变,并对肾损害进行半定量评分。结果:糖肾方可降低模型组大鼠血中TNF-α、DA水平,升高IL-10水平。降低肾小球硬化指数和肾小管间质纤维化指数,改善肾组织形态学异常。结论:糖肾方对2型糖尿病模型OLETF大鼠肾脏具有保护作用,延缓其病理进展,其作用机制可能与调节体内炎症因子水平有关。
Objective:To observe the renal protective effect of Tangshen Formula(TF) in the spontaneous rat model of type 2 diabetic nephropathy-OLETF rats and explore the possible molecular mechanism. Methods:OLETF rats were randomly divided into three groups: model group, TF group(1.6 g/kg) and monopril group(1.0 mg/kg).LETO rats with the same week age and sex were used as control group. Tangshen Formula was given to the relevant group after 12 week for 44 weeks by intragastric administration. Levels of blood glucose and 24 hours urinary protein were determined every 4 weeks. When the trial was finished, blood samples were taken from the abdominal aorta and TNF-α, IL-10 and DA in blood serum were detected. Kidney tissue was taken for histopathological and pathological change of glomeruli and interstitial of nephric tubule were observed by semiquantitative assessment. Results:Comparing with the model group, TF group decreased the levels of TNF-α and DA, as well as golmerulosclerossi index and tubular fibrosis index, increased the level of IL-10. Conclusion:Tangshen Formula has a good renal protective effect in rats with diabetic nephropathy. The mechanism may be partly related to inhibiting the inflammatory cytokine expression.
