目的探究常规实验操作对动物福利的损伤程度以及音乐干预的效果。方法采用3周龄雄性ICR小鼠,实验操作组分别给予保定(E-r)、灌胃(E-g)、腹腔注射(E-p)和眼眶静脉丛采血(E-o)处理,音乐干预组采取相同处理后给予1h音乐干预(M-r,M-g,M-p,M-o)。连续处理7 d后,测定与福利相关的神经内分泌、免疫、生化、生长指标和饲料能效比。结果 ①与对照组相比,实验操作四组体增重和饲料能效比均显著降低;E-r组TG、HDL-C、WBC、IL-2、β-EP和脾脏、胸腺指数均显著降低,CORT、ACTH、EPI显著升高;E-g组的TG、WBC、IL-2和β-EP显著降低,CORT、ACTH和EPI显著升高;E-p组IL-2显著降低,CORT、ACTH和EPI显著升高;E-o组脾脏指数和WBC显著降低,CORT显著升高。②与实验操作组相比,M-r组体增重、饲料能效比、脾脏指数、IL-2显著升高,CHOL、CORT、ACTH和β-EP均显著降低;M-g组WBC显著升高,CORT显著降低;M-p组WBC显著降低;M-o组饲料能效比显著升高。结论四种常规实验操作均对小鼠的健康福利造成了不同程度损伤,而音乐干预能够改善小鼠的操作应激和福利损伤程度。
Objective To study the impaired-welfare induced by routine experimental procedures on mice and the effect of music intervention. Methods 3week male ICR mice were divided into 9 groups. The experimental groups were treated by several experimental operations including restraint( E-r),intragastric injection( E-g),intraperitoneal injection( E-p) and orbital venous sampling( E-o). The music intervention groups were given 1 hour music intervention after the same experimental operation( M-r,M-g,M-p,M-o). After 7 days of continuous treatment,the main neuroendocrine,immune,biochemical parameters,body weight and food efficiency ratio( FER) were measured. Results ① Compared with the untreated group,the body weight gain and FER were significantly decreased in such four experimental groups; TG,HDL-C,WBC,IL-2,β-EP,spleen index and thymus index were significantly decreased in the E-r group,in addition,CORT、ACTH and EPI were significantly increased; TG,WBC,IL-2 and β-EP were significantly decreased in the E-ggroup,and CORT,ACTH,EPI were significantly increased; IL-2was significantly decreased in the E-p group,and CORT,ACTH,EPI were significantly increased; The spleen index and WBC were significantly decreased in E-o group,and CORT was significantly increased. ② Compared with the experimental groups,the body weight gain,FER,spleen index,IL-2were significantly increased in the M-r group,and CHOL,CORT,ACTH,β-EP were significantly decreased; WBC was significantly increased in the M-g group,and CORT was significantly decreased; WBC was significantly decreased in the M-p group; FER was significantly increased in the M-o group. Conclusion The four routine experimental procedures cause strong stress response in mice,and lead to health and welfare impaired. However,music intervention played an important role in stress injury improvement.
