目的:观察眼针对脑缺血再灌注损伤(CIRI)模型大鼠脑皮层组织TNFR1表达的变化,探讨眼针治疗CIRI的作用机制。方法:健康雄性SPF级Wistar雄性大鼠60只,随机分为正常组、假手术组、模型组和眼针组。模型组和眼针组采用改良的线栓法建立大鼠大脑中动脉缺血(MACO)再灌注动物模型。分别以免疫组化和RT-PCR方法观察缺血侧脑皮层TNF-α、TNFR1蛋白表达,TNFR1 mRNA表达的变化。结果:与正常组及假手术组比较,模型组大鼠缺血侧脑皮层TNF-α、TNFR1的蛋白及基因均显示出明显的高表达(P〈0.01),而眼针组同模型组相比,TNF-α、TNFR1表达均下调(P〈0.05)。结论:眼针抑制CIRI大鼠TNF-α、TNFR1的高表达,可能是眼针治疗CIRI的作用机制之一。
Objective:To evaluate the effect of eye acupuncture resisting acute focal cerebral ischem/ reperfusion(CIRI) injury in rats,and to explore the mechanism through signal t ransduction pathway.Methods:Adult male SPF Wistar male rats were randomly divided into normal group,sham operation group,CIRI model group and the eye acupuncture group.There were 15 rats in every group.The model of middle cerebral artery occlusion(MACO) was established by using improved suture method.Immunohistochemical method was performed to examine the protein expression of TNF-α and TNFR1 in the ischemic brain cortex;RT-PCR methods was performed to examine the mRNA expression of TNFR1 the ischemic brain cortex.Results:The protein expression of TNF-α and TNFR1 in CIRI model group were increased significantly(P0.01) compared with those in normal group and sham operation group,and which were decreased significantly(P0.05 or P0.01) in eye acupuncture group;RT-PCR results showed that compared with the normal group and sham operation group,mRNA expression of TNFR1 in model group ischemic brain cortex was significantly increased(P0.01),which was significantly lower expression in eye acupuncture group(P0.01).Conclusion:Eye acupuncture therapy shows protective effects on CIRI partly through intervening TNF-α/TNFR1 signal transduction pathway.eye acupuncture therapy reduce the incidence of apoptosis,and play a neuroprotective role,which may be eye acupuncture therapy for CIRI's part of the treatment mechanism.
