目的:研究栀子配方颗粒对正常昆明小鼠血脂及肝毒性的影响。方法:连续灌胃28d,观察小鼠行为形态学变化;测定小鼠体重与体长,计算Lee's指数;测定小鼠肝脏湿重并计算肝脏指数;测定血清中TG(甘油三酯)、TC(总胆固醇)、AST(天门冬氨酸氨基转移酶)的含量;对肝脏组织进行病理学检查。结果:栀子配方颗粒浓度1g/kg、3g/kg、6g/kg、9g/kg、12g/kg、16g/kg组的TG均显著降低;血清中TC含量与空白组相比无显著差异;栀子浓度16g/kg组的体长显著增大,Lee's指数则明显降低;栀子浓度16g/kg组的肝脏指数、AST含量显著升高,与空白组比较P〈0.05;光镜下栀子浓度16g/kg组的肝细胞可见明显的肿胀坏死、炎细胞浸润等形态学上的改变。结论:栀子具有显著的降低血清TG作用,但对TC作用不显著;栀子能增加小鼠体长,降低Lee's指数,16g/kg组作用尤为显著;长期低剂量服用栀子对肝脏无影响,但长期大剂量服用则易造成肝毒性。
Objective: To study the lipid-lowering and hepatotoxicity effect in mice of the Gardenia formula particle. Method: Concoct the Gardenia formula particles into different concentration of solution with distilled water. Observe the change of the mice behavior,weight and other index through oral gavage daily for 28 d. Measure mice weigh and body length and calculate the Lee’s index. Weight the liver and calculate the liver index. Detect the AST,TC and TG. Observe the liver tissue by optical microscope. Result: During the treatment,every group had a slowly growth on weight,and the mice back appeared slightly depilation phenomenon. And all of the mice were doing well in mental state.Detect serum TG and TC found that Fructus Gardenia had a significantly effect on lower serum TG,but had no obvious effect on serum TC.Gardenia can increase the mice body length and reduce the Lee ’s index,compared to the blank group,the Gardenia 12 g / kg,16 g / kg dose group had a significantly effect on it. Compared to the blank group,the serum AST level of the Gardenia solution of the 1g / kg,3g / kg dose group were slightly reduced,and the 6g / kg、9g / kg dose group were slightly higher,but the 12 g / kg,16 g / kg dose group were obviously higher. Weight the liver and calculate the liver index,and compared with the blank group,Gardenia 1g / kg,3g / kg dose group were decreased;Otherwise,the 16 g / kg dose group had a significant rise in the liver index. It was also found that the liver cells of the Gardenia1 g / kg,3g / kg dose group had not remarkable different from the blank group,and the 16 g / kg dose group were obviously swell,necrotic and changed with inflammatory infiltrate through the optical microscope. Conclusion: Fructus Gardenia long-term low dose taken had no toxicity to the liver,and had the effect of decreased the blood fat. If the dose was too excessive,it would cause liver damage.
