目的:研究转移抑制基因KAI1对人鼻咽癌细胞体外增殖、侵袭及转移能力等生物学效应的影响。方法:通过腺病毒载体将KAI1基因转染人鼻咽癌细胞株HNE-1,RT-PCR法检测转染效果;MTT法检测鼻咽癌细胞转染KAI1基因后体外增殖能力的变化;FCM法检测细胞生长周期分布;Transwell侵袭小室检测细胞侵袭能力的改变;细胞同质黏附实验检测细胞黏附力的变化。结果:当腺病毒载体rAd-KAI1效价为30 MOI时,对HNE-1细胞的转染率可达92%;转染48和72 h时,rAd-KAI1转染组细胞的增殖明显受到抑制,抑制率分别为17%和30%;G0/G1期细胞数量较对照组增多,S期细胞数量减少;转染组的穿膜细胞数为(52.5±4.2)个,明显低于对照组的(167.4±3.5)个,差异有统计学意义(P〈0.05);转染组的细胞同质黏附作用高于对照组(P〈0.05)。结论:腺病毒载体成功介导了抑癌基因KAI1转染进入鼻咽癌HNE-1细胞,并取得较高的转染率。KAI1基因可能通过阻滞HNE-1细胞生长周期,从而抑制细胞增殖。KAI1基因能够抑制HNE-1细胞侵袭能力,其机制可能为KAI1基因增强了鼻咽癌细胞的黏附力所致。
Objective:To study the effect of metastasis suppressor gene KAI1 on the proliferation,invasion and metastasis of nasopharyngeal carcinoma HNE-1 cell line.Methods:The KAI1 gene was transferred into human nasopharyngeal carcinoma HNE-1 cell line via adenovirus mediation.The transfection efficiency of KAI1 gene was measured by RT-PCR.The effect of KAI1 gene on the proliferation of HNE-1 cell line was tested with MTT assay.The distribution of cell cycle was determined by flow cytometry(FCM) analysis.The invasion ability of the HNE-1 cell lines was determined by using Transwell chamber.The adhesion capability of KAI1-transfected HNE-1 cells was detected by using microorganism adhesion test.Results:When the concentration of the adenovirus was 30 MOI,the transferred rate of HNE-1 cell line was above 92%.MTT assay showed that the proliferation of rAd-KAI 1 transfected HNE-1 cells was inhibited by 17% at 48 h and 30% at 72 h compared to control group(P〈0.05).The proportion of cells in G0/G1 phase was increased and that in S phase was decreased.The invasion assay demonstrated that the number of HNE-1 cells crossing the membrane in KAI1 transfected group was less than that in the non-transfected group [(52.5±4.2) vs(167.4±3.5,P〈0.05)].The homogeneous adhesion of KAI1-transfected HNE-1 cells was higher than that in control group(P〈0.05).Conclusion:The adenovirus successfully mediated the transfection of tumor suppressor gene KAI1 into human nasopharyngeal carcinoma HNE-1 cells with higher transfection rate.KAI1 gene inhibited the growth of HNE-1 cells by arresting the cell cycle at G0/G1 phase.KAI1 gene reduced the invasion ability of HNE-1 cells by enhancing the adhesion capability of HNE-1 cells.
