目的:观察川红拔瘤膏外敷对小鼠G422脑胶质瘤生长的抑制作用及其抗肿瘤的机制。方法:选取清洁级小鼠50只,随机分为:对照组、环磷酰胺组、川红拔瘤膏高、中、低3个剂量组。中药组右腋下皮肤涂抹给药,给药量为0.5mL。连续给药14天,阳性对照组连续给药7天。连续观察川红拔瘤膏对肿瘤生长的抑制情况,末次处死后,取肿瘤组织,流式细胞仪检测细胞周期变化与细胞凋亡。RT-PCR法检测肿瘤组织中caspase-3的表达。结果:拔瘤膏高浓度组的瘤重(0.516±0.107)g与其他组比较(除环磷酰胺组)对实体瘤的生长具有显著的抑制作用(P〈0.05)。高浓度组肿瘤抑制率为31.28%。流式细胞仪结果显示,与空白组比较川红拔瘤膏高剂量组明显将小鼠肿瘤组织细胞阻滞于G0/G1期(P〈0.05),S期细胞数明显减少(P〈0.05)。细胞凋亡分析可知,高剂量组作用的肿瘤细胞的凋亡率为(23.6±4.35)%,明显高于对照组(8.86±1.23)%,差异有统计学意义(P〈0.05)。其他组与对照组比较,差异无统计学意义。caspase-3分析结果显示,与低剂量组、中剂量组,caspase-3表达强度无明显改变[(8.23±0.43),(7.96±0.52)vs(7.24±0.36),P〉0.05]。高剂量组的caspase-3表达显著增加[(23.45±2.31)vs(7.24±0.36),P〈0.01]。结论:川红拔瘤膏对小鼠G422脑胶质瘤具有抑制生长的作用,其作用可能与其抑制肿瘤细胞DNA合成、周期阻滞、诱导凋亡及上调凋亡基因caspase-3表达有关。
Objective:To investigate the effect of Chinese medicine Recipe by external application for detecting the rat tumor growth inhibition,then explore the anti-tumor mechanism of action.Methods:Fifty mice were divided into 5groups,the normal group,cyclophosphamide group,high dose of plucking tumor mastic group,medium dose of plucking tumor mastic group,low dose of plucking tumor mastic group.Three groups of treatment by plucking tumor mastic(high dose,medium dose,low dose)were given 0.5ml dose respectively by smearing skin under right armpit for 14 days and cyclophosphamide group was given cyclophosphamide for 7 days,in order to observe continuously the condition of rat tumor growth inhibition by treatment of plucking tumor mastic.From tumor tissue in rat,cell cycle and number of apoptotic cells were detected by flow cytometer,and expression of caspase-3 was determined by reverse transcription polymerase chain reaction method.Results:In G422 cell of rat model,except for cyclophosphamide group,high dose of plucking tumor mastic group was obviously growth inhibition when compared with other groups,which its rate of growth inhibition was 31.28%.According result of flow cytometer,high dose of plucking tumor mastic group made obviously tumor tissue cells stop stage of G0/G1(P0.05),and cells in stage of S decrease(P0.05).No significant difference between the normal group and the other groups,but the rate of apoptotic cells in high dose of plucking tumor mastic group was (23.6±4.35)% from apoptotic cells analysis,which was obviously higher than the normal group(8.86±1.23)%.With dose gradually increasing,expression of caspase-3 of high dose of plucking tumor mastic group was significantly superior to other groups[(23.45±2.31)vs(7.24±0.36),P0.01].Conclusion:Chinese medicine Recipe could inhibit the tumor growth on rat's model.The anti-tumor mechanism of action was about related to inhibit the composition of tumor cell DNA,stoppage cycle,induce the apoptosis of G422 cells and up-regulat
