目的研究六味地黄丸对胃癌细胞及人胃癌裸鼠移植瘤的作用。方法制备六味地黄丸含药血清,观察在含药血清作用下的BGC-823胃癌细胞生长情况,MTT法检测各组细胞增殖抑制率。建立人胃癌裸鼠移植瘤模型,分为对照组、六味地黄丸低剂量组、中剂量组、高剂量组、5-Fu组、六味地黄丸低剂量+5-Fu组、中剂量+5-Fu组、高剂量+5-Fu组。六味地黄丸灌胃,观察裸鼠一般情况,1周后处死荷瘤鼠,称重瘤块并计算抑瘤率。HE染色,观察各组胃癌细胞坏死情况。结果给药48 h后,六味地黄丸中剂量+5-Fu组的胃癌细胞存活率显著低于对照组,差异具有统计学意义(P〈0.01)。药物作用后与对照组比较,5-Fu组、六味地黄丸低剂量+5-Fu组、中剂量+5-Fu组、高剂量+5-Fu组的抑瘤率分别为54.250%、57.873%、73.540%、72.625%;且该4组裸鼠体重均较对照组降低。单纯5-Fu组比六味地黄丸+5-Fu组腹泻严重,差别有统计学意义。结论六味地黄丸联合5-Fu对抑制胃癌细胞增殖有协同作用,且增强裸鼠体质。
[ Objective ] To investigate the effect of Liuweidihuangwan on the gastric cancer cell and the human gastric carcinoma xenograft in nude mice. [ Methods ] Made Liuwei Dihuang wan drug serum, the growth of BGC- 823 gastric cancer cell was observed after the drug serum. MTT was used to investigate the inhibitory effect of caner cells. Establish the model of human gastric carcinoma xenograft in nude mice, model mice were randomly divided into blank group, Liuweidihuangwan-low(LLD) group, Liuweidihuangwan-mid(LMD) group, Liuweidihuangwan-high(LHD) group, 5-Fu group, liuweidihuangwan-low+5-Fu(LLD+5-Fu) group, Liuweidihuangwan-mid+5-Fu (LMD+5-Fu) group, Liuweidihuangwan-high+5-Fu (LHD+5-Fu) group. Nude mice were treated by intragastric administration of Liuweidihuangwan, the general conditions of nude mice were observed. Nude mice were executed after one week, then the xenografts were weighted and the anti-tumor rate was calculated. Hematoxylin eosin staining(HE)was used to detect the death of cancer cell in tumor tissue. [ Results ] The viability of gastric cancer cells of LMD+5-Fu group was significantly lower than that of the blank group after being administrated by Liuweidihuangwan and 5-Fu for 48h. The difference was statistically siginificant (P〈0.01). Compared with the blank group, the anti-tumor rate of the 5-Fu group was 54.250%, the anti-tumor rate of the LLD+5-Fu group was57.873%, the anti-tumor rate of the LMD+5-Fu group was 73.54%, the anti- tumor rate of the LHD+5-Fu group was 72.629%,and the weight of nude mice in the four groups were lost compared with the blank group. The symptom of scour in 5-Fu group was significantly more serious than that of 5-Fu+LD group during the therapeutic process. [ Conclusion ] Liuweidihuangwan and 5-Fu have synergistic effect on controlling the growth of the gastric cancer cell and enhance the habitus of nude mice.
