目的探讨PARP-1抑制剂3-AB对脓毒症大鼠心肌细胞PARP-1、Bcl-2表达的影响。方法成年健康SD大鼠30只,随机分为3组:假手术对照组、模型组和3-AB治疗组。采用盲肠结扎穿孔术制作脓毒症大鼠模型,存活至48h取材。用免疫组织化学sP法和图像分析技术检测心肌PARP-1、Bcl-2的表达,ELISA法检测血清中PARP-1、Bcl-2的含量。结果模型组心肌PARP-1阳性细胞灰度值、血清中的OD值(opticaldensity)均较假手术组升高,而治疗组较模型组下降,差异有统计学意义,P〈0.05;而Bcl-2的变化趋势正好相反,模型组心肌Bcl-2阳性细胞灰度值、血清中的OD值均较假手术组降低,3-AB治疗后其水平较模型组升高,P〈0.05。结论PARP-1抑制剂3-AB可上调脓毒症大鼠大心肌Bcl-2的表达,对凋亡有-定的抑制作用。
Objective To explore the effect of 3-AB on expression of PARP-land Bcl-2 in myocardium of rats with sepsis. Methods 30 adult heahh rats were randomly divided into the sham operation group, the model group, and the therapy group with 3-AB. The rats were sacrificed on 48 hours after operation. The expression of PARP-1 and Bcl-2 in myocardium was determined by immunohistochemical staining. The immunepositive gray scale of the immune reactant was detected by computer image analysis technique. The OD (optical density) of PARP-1 and Bcl-2 in serum were detected by ELISA. Results In the model group, the expression of PARP-1 in the myocardium increased apparently compared with the sham operation group, P 〈0.05, but it decreased after therapy with 3-AB , P 〈0.05. Conversely, the expression of Bcl-2 in the myocardium decreased apparently in the model group after operation compared with the sham operation group, P 〈0.05, but it increased markedly after therapy with 3-AB compared with the model group, P 〈0.05. Conclusion The inhibitor of PARP-I (3-AB) could up-regulate the expression of Bcl-2 in myocardium of rats with sepsis and has inhibitive effect of apoptosis.
