细菌脂多糖(lipopolysaccharide,LPS)可激活单核/巨噬细胞、内皮细胞合成和释放多种细胞因子,导致全身性炎症反应.LPS识别及跨膜信号转导是引起细胞效应的关键.在过去的几年中,有关LPS结合蛋白家族和受体系统及其作用机制的研究突飞猛进,大量研究表明LPS识别涉及复杂的蛋白质间相互作用.在此对LPS识别系统成员及其功能的最新研究进展进行综述,并详细介绍新近提出的“LPS受体激活簇”理论.
Bacterial lipopolysaccharide (LPS) induces cytokine synthesis and secretion in monocytes/ macrophages and endothelial cells, resulting in subsequent inflammatory responses of multiple systems. The receptor recognition of LPS and the transmembrane signal transduction play a key role in LPS-induced cell activation. In recent years, the intensive studies on analyzing LPS receptors and their associated proteins have suggested the molecular mechanism of LPS recognition involves complicated protein-protein interactions. In this review, a novel theory of LPS receptor activation cluster will be introduced, in addition, the recent advances in the mechanistic studies of LPS receptor system and its components will be discussed.
