目的观察视网膜Müller细胞中碱性成纤维细胞生长因子(bFGF)在豚鼠形觉剥夺性近视(FDM)中的表达,进一步探讨近视眼的发病机制。方法用半透明眼罩遮盖法建立豚鼠FDM模型;采用玻璃体腔注射L-α-氨基己二酸(L-α-AAA)破坏视网膜Müller细胞,观察其对豚鼠FDM形成的影响。视网膜检影验光测屈光度,A型超声测量眼轴长度。免疫组织化学法观察bFGF在视网膜Müller细胞中的表达。结果遮盖眼与对照眼相比,产生了相对性近视(P〈0.05),眼轴增长(P〈0.05);去遮盖后,近视度数降低(P〈0.05);破坏视网膜Müller细胞后,近视度数较单纯遮盖眼降低(P〈0.05)。近视眼与对照眼相比,视网膜Müller细胞中bFGF表达减弱。结论视网膜Müller细胞可能参与了FDM形成的调控;视网膜Müller细胞中bFGF的表达可能与FDM有关。
Objective The expression of basic fibroblast growth factor(bFGF)in retinal Müller cell of form deprivation myopia(FDM)in guinea pig was observed in present study for exploring the pathogenesis of FDM.Methods FDM model was established on the right eyes in 40 guinea pigs by wearing the semitransparent blinders for 2 weeks.FDM models were divided into 4 experimental groups.Stable wearing of semitransparent blinders was selected in the stable covering group.The fashion of two-week shading and 1-week opening was selected in the covering removal group.The right eyes were shaded for 2 weeks and 10 μL L-α-aminoadipic acid(L-α-AAA)was vitreously injected before and 1 week after modeling in the L-α-AAA group.The right eyes were shaded for 2 weeks and PBS was vitreously injected at a same way in PBS group.Ten normal guinea pigs were as control group.Refractive status and axial length of experimental eyes and control eyes were measured with retinoscopy and A-scan ultrasonography.The expression level of bFGF in the Müller cell of retina was analyzed using immunohistochemistry.Results The significant differences were found in refractive diopter of FDM and axial length in different groups(F=17.697,P=0.004 7;F=16.863,P=0.005 9,respectively),indicating that the refraction diopter and axial length were increased in comparison with normal control group(P〈0.05).The morphology of Müller cell was identified by GFAP staining,showing brown staining between outer limit membrane and inner limit membrane,and bFGF was expressed in multipel layer of retina with the lowest tensity of expression in normal control group compared with other various groups(P〈0.05).Refractive status of experimental eyes was reduced relatively after removeing blinder(P〈0.05),showing that the expression of bFGF in FDM eyes was down-regulated in comparison with control eyes.Conclusion The Müller cells of retina may be involved in the regulation of FDM formation.Expression of bFGF in the Müller cells may be related to FDM.
