目的研究视网膜Maller细胞在豚鼠形觉剥夺性近视形成中的作用。方法眼罩遮盖建立豚鼠形觉剥夺性近视眼模型,分为正常对照组、DL—α-氨基己二酸(DL—α—AAA)组、生理盐水组、遮盖组、遮盖+DL—d—AAA组、遮盖+生理盐水组。DL-α-AAA组和遮盖+DL-α—AAA组右眼玻璃体内注射8μgDL—α—AAA。14d后检影验光测屈光度,A型超声测眼轴长度,免疫组织化学、Western blotting检测视网膜中Vimentin的表达,TUNEL染色检测凋亡细胞。结果眼罩遮盖14d后,豚鼠遮盖眼眼轴延长,近视形成。Vimentin蛋白阳性表达于视网膜MUller细胞。DL—α-AAA组右眼远视度数高于正常对照组,但视网膜Vimentin蛋白表达量下调(P〈0,05)。DL-α-AAA引起遮盖眼近视度数减轻、视网膜Vimentin蛋白表达量下调(P〈0.05)。各实验组的视网膜均未发现凋亡细胞。结论玻璃体内注射DL—α-AAA能特异性作用于视网膜Mtiller细胞,有效抑制豚鼠眼球正视化和近视形成。
Objective Present study was to investigate the effect of retinal Muller cells on form deprivation myopia in guinea pig. Methods Form deprivation myopia in guinea pig was induced by occlusion of the right eyes with translucent goggles. The guinea pigs were divided into 6 groups:normal control group, DL-alpha-aminoadipic acid (DL-α-AAA) group, saline solution group,deprivation group,deprivation + DL-α-AAA group and deprivation + saline group. 8 μg DL-α-AAA was injected into the vitreous chamber of the right eye of guinea pig in DL-α-AAA group and deprivation + DL-α-AAA group. After 14 days of goggles occluding, myopia was confirmed by optometry and A ultrasonography. Subsequently, retinal Vimentin protein was evaluated by the immunohistochemistry and Western blotting,and retinal apoptotic cells were detected by TUNEL. Results Form deprivation myopia formed in occluded eyes after 14 days. Vimentin protein was positively expressed in retinal Miiller cells of guinea pig. Hyperopic diopters of the right eyes was higher in DL-α-AAA group and accompanied by the decrease of retinal Vimentin protein in comparison with normal control group ( P 〈 0.05 ). The injection of DL-α-AAA into vitreal chamber caused a marked decrease in myopic diopters and downregulation of retinal Vimentin protein ( P 〈 0.05 ). No apoptotic retinal nuclei was detected in all groups. Conclusion The intravitreal DL-α-AAA specifically acts on retinal MUller cells, and effectively ameliorates emmetropization and myopic ocular growth in guinea pig.
