针对微量三磷酸腺苷(ATP)检测需求,利用生物荧光技术,设计了一种手持式ATP检测仪。该检测仪以小型光电倍增管PMT为光电转换器件,通过有效的干扰屏蔽和信号采集放大电路设计,实现了ATP反应释放微弱荧光信号的采集。以10^-15-10^-9mol/L标准ATP样品为检测对象,以大型分析仪器F4500为对比仪器,进行了梯度对比实验,结果表明,本仪器可检测最低ATP浓度为10^-14mol/L,是F4500最低检测限的1/100。检测时间为10s时,在10v-14-10^-9mol/L浓度范围内,本仪器响应值与标准ATP样品浓度在对数坐标下线性相关,相关系数达到了0.9902。
A handheld Adenosine Triphosphate (ATP) detecting system based on bioluminescence was described. In the system, a small photomultiplier tube (PMT) module was used to measure the weak bioluminescence released from ATP reaction. The system was used to detect ATP in the range of 10^-15-10^-9 mol/L. The results were compared to that of fluorescence spectrophotometer F4500 which showed the lowest ATP concentration the system could detected was 10^-14mol/L which was 1/100 of that F4500 could do. In 10s' time, the response signals correlated well to the ATP concentration in the range of 10^-14- 10^-9 mol/L with the correlation coefficient of 0. 9902.
