目的:使用补肾通脉汤对兔动脉粥样硬化(atherosclerosis,AS)造模过程进行干预,通过对AS斑块内基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和其基质金属蛋白酶抑制物-2(tissue inhibitor of metalloproteinase-2,TIMP-2)表达的进行检测,探讨补肾通脉汤阻碍AS进展、稳定AS斑块的机制。方法:选取健康新西兰雄性大耳白兔40只,随机分成4组,空白对照组(CG),模型组(MG),补肾通脉组(BSG),阿托伐他汀组(ACG),每组10只,高脂饲料喂养及牛血清白蛋白耳缘静脉注射制作动脉粥样硬化模型,免疫组化法检测斑块内MMP-2和TIMP-2在斑块内的表达的阳性细胞面积。结果:与CG组比较,MG组、BSG组、ACG组MMP-2和TIMP-2表达均有显著升高(P〈0.001);与MG组比较,BSG组、ACG组MMP-2和TIMP-2表达均有显著降低(P〈0.001,P〈0.01),BSG组与ACG组MMP-2和TIMP-2表达无明显区别(P〉0.05)。结论:补肾通脉汤可以通过调节MMP-2和TIMP-2在兔AS模型斑块内的表达,有效阻碍AS进展、稳定AS斑块。
Objective:To investigate the mechanisms that affects AS progress and plaque vulnerability through detecting the expression of MMP-2(matrix metalloproteinase-2,MMP-2)and TIMP-2(tissue inhibitor of metalloproteinase-2,TIMP-2)in AS plaques by intervening with Bushentongmai decoction in the model of AS rabbits.Methods :40 male rabbits of New Zealand were divided into two groups at random,CG(control group,CG),MG(model group,MG),BSG(group of bushentongmai decoction,BSG),ACG(group of atorvastatin calcium,ACG),and there are 10 rabbits in every groups.To make AS model by high lipid diet and injecting the vein of ear fringe with BSA(bovine serum albumin,BSA).Analyze the expression of MMP-2 and TIMP-2 in AS plaques by HIS(immunol histochemistry staining,HIS).Result :The expression of MMP-2 and TIMP-2 increased significantly of the other three groups compared with CG(P0.001).The expression of MMP-2 and TIMP-2 decreased significantly in BSG,ACG compared with MG(P0.001,P0.01).There is no difference of the expression of MMP-2 and TIMP-2 between BSIG and ACIG(P0.05).Conclusion :Bushentongmai decoction can regulate the expression of MMP-2 and TIMP-2 in the AS plaques,obstruct the process and stable the AS plaque.
