目的:用体质量监测、糖水消耗、行为学方法评价逍遥散提取物对肝郁脾虚证模型大鼠的疗效。方法:运用复合提取法将逍遥散原方分离为A、B、C三种组分,对慢性束缚应激大鼠模型进行干预。21d造模给药结束后测量大鼠体质量、糖水偏好百分比、行为学指标等,评价各提取物的疗效。结果:与模型组相比,逍遥散提取物中剂量组能改善大鼠体质量的增长(P〈0.05),增加糖水消耗百分比(P〈0.05)及旷场站立次数(P〈0.01),降低新环境进食探索时间(P〈0.05);提取物组分C能增加旷场运动总长度(P〈0.01),降低新环境进食探索时间(P〈0.05)。结论:逍遥散提取物中剂量及组分C对肝郁脾虚证模型大鼠有较好的疗效,适用于后续研究。
Objective: To evaluate the effect of Xiaoyao San extract in regulating rat model with stagnation of liver qi and spleen deficiency syndrome through weight monitoring, sucrose preference test and behavioral methods. Methods: We divided Xiaoyao San prescription into three different fractions (XYS-A, XYS-B, XYS-C) with multiple extraction methods, used them to regulate the rat model with stagnation of liver qi and spleen deficiency syndrome subjected to chronic immobilization stress (CIS). After 21 days' modeling and administration, the body weight, sucrose preference, open-field test sores and latency of novelty suppressed feeding test (NSF) were measured to evaluate the effect of Xiaoyao San extract. Results: Compared with the model group, the middle dose group of fractions mixing could increase the rats' body weights (P〈0.05), the percentage of sucrose consumption (P〈0.05), the number of rearings (P〈0.01) and decrease the latency time of feeding (P〈0.05). Compared with the model group, the XYS-A group could increase the total length of movement in open-field (P〈0.01) and decrease the latency time of feeding (P〈0.05). Conclusion: The middle dose group of fractions mixing of Xiaoyao San and XYS-A have good effects in regulating rat model with stagnation of liver qi and spleen deficiency syndrome, which are suitable for the follow-up study.
