目的:观察liguzinediol(2,5-二羟甲基-3,6-二甲基吡嗪)对戊巴比妥钠致急性心力衰竭大鼠血流动力学的影响。方法:40只雄性SD大鼠右侧股静脉恒速注射戊巴比妥钠制备急性心衰模型,造模后随机分为5组:模型组(给予NS),阳性对照组(地高辛,22.5μg·kg-1),liguzinediol(5,10,20mg·kg-1)3个剂量组,每组8只,稳定后经恒速注射泵右侧股静脉单次给药,多道生理记录仪记录全过程中大鼠血流动力学指标的变化。结果:静脉给药后,liguzinediol3个剂量组均使大鼠左心室收缩压(LVSP)、左心室内压力最大上升速度(+dp/dt max)、左心室内压力最大下降速度(-dp/dt max)、收缩压(SAP)、舒张压(DAP)在1min内开始显著增加,并且对心率影响不大,心肌收缩力增强。结论:liguzinediol对急性心衰大鼠心脏具有正性肌力作用。
Objective: To investigate the effect of liguzinediol on the hemodynamics in acute heart failure rats. Method: Forth rats were intravenously injected pentobarbital through a micro-infusion pump and randomly divided into 5 groups: saline control group (NS) , positive control group (digoxin, 22.5 μg· kg-1), three doses of liguzinediol groups (liguzinediol 5, 10, 20 mg . kg-1). The heart contractile founctions such as left ventrieular systolic pressure (LVSP) , maximal rising and falling rate of ventricular pressure ( + dp/dtmax ) , systolic arterial pressure (SAP) , diastolic arterial pressure (DAP) , heart rate (HR) were measured using a RM6240B/C four channel physiological recording instrument. Result: Liguzinediol induced significant increases of LVSP, + dp/dt -dp/dt and SAP. Conclusion: Liguzinediol induces a positive inotropic effect on the heart in acute heart failure rats.
